[Objecti ve] The normal and degenerative intervertebral disc nucleus pulposus cells (Nucleus pulposus of intervertebral disc) type II collagen staining and safranin staining were compared with "O";.[Method] The removal of voluntary patients were obtained from 3 patients with scoliosis and 3 cases of protrusion of intervertebral disc nucleus pulposus in waste, the measurements of 26 cells were cultured in each patient, the normal group and the degeneration group each measuring 78 cells. Type II collagen in normal and degenerative intervertebral disc nucleus pulposus cells were observed by immunofluorescence staining and quantitative determination,; nucleus pulposus cells for some values of "O" red staining and gray.[Result] A. staining:degeneration of the intervertebral disc nucleus pulposus cells were spindle shaped, oval, spindle andirregular in shape, cell staining light, fuzzy, visible only in very small amounts of intracellular fluorescent particles;expression of collagen type II decreased significantly (P<0.05); "O" B. safranin dye color:degenerative nucleus pulposus thin. Cell morphology was irregular, nuclear shape is irregular, weak staining, staining cell protrusions andcytoplasm is not uniform, decomposition structure parts of cells, loss of cell integrity, disorganized, sparse, some may be observed in a few cells necrosis; image gray value compared with no difference (P>0.05).[Conclusion] The normal and degenerative intervertebral disc nucleus pulposus cells, degeneration of nucleus pulposus cells decreased, and even apoptosis, type Ⅱ collagen content in the cell is also reduced significantly.
