Objective:American cockroach (Periplaneta americana) plays an important role in the pathogenesis of asthma. Preparation of high quality allergen extracts is essential for the diagnosis and immunotherapy of allergic disorders. Our aim was to develop a standardized procedure to prepare American cockroach extract (ACE).Methods:We evaluated the influences of various parameters, such as extraction time, sodium chloride (NaCl) concentration, pH in sample solutions on the extraction efficiency. In addition the procedure was then optimized using orthogonal test. Also, the major allergens in the extract were examined by immunoblot analysis.Results:Ranking of extract potency was NaCl concentration, extraction time followed by pH. The NaCl concentration in sample solution significantly impacted the yield of ACE (P<0.05). The optimized condition for ACE extraction was as follows:NaCl concentration 0.4 mol/L, extraction time 6 h and pH 8.0.Conclusion:The ACE was prepared in a standardized way, which will be useful for the diagnosis and management of allergic respiratory diseases. |
