Ginseng is dry root and rhizome of Panax ginseng C. A. Meyer, which contains a variety of chemical components, such as ginsenosides, polysaccharide, protein, amino acids, flavonoids, inorganic elements, vitamins, organic acids, alkaloids, fat, sterol, nucleoside, lignin, volatile oil and other substances. More ginseng grow in 40° to 45° north latitude and 117.5° to 134° east longitude, densely in Liaoning province, Jilin province, part of the eastern of Heilongjiang province, and a little part of Shanxi, Shandong and Hebei province is also sown. South Korea, North Korea, the Soviet Union and Japan are also more cultivated.Ginseng has many biological activities and pharmacological effects, such as anticancer, anti-aging, improving immunity, reducing blood glucose, decreasesthe level of blood-lipid and increasing angiogenesis, and so on. Modern medicine studies have shown that ginseng protein and amino acids also played an important role in the human growth, development, health care and anti-aging. Therefore, the effect of ginseng protein and amino acids are also not allow to ignore. Ginseng is rich in protein. There are so many methods of its extraction, separation and determination. But in recent years, Coomassie Brilliant Blue(CBB) has became an essential toll of the content determination of protein in Traditional Chinese Medicine(TCM). Currently reports on the analysis of the composition of amino acids in the ginseng are more, but mostly relies on the automatic amino acid analyzer for the determination of total amino acids. There is no found that someone use HPLC-ELSD to determinate the content of the amino acids in ginseng. Ginseng also contains organic acids and nucleoside compositions, and so on.This paper studied the content of ginseng protein, amino acids, organic acids and nucleotide compositions from the different regions 42 ginseng samples in different years.Use Tris-HCl to extract and use polyethylene glycol 6000 to precipitate, and according to the CBB method to determinate the content of protein in ginseng.Use HCl as solvent to hydrolyze ginseng under the condition of 110℃ for 24 h, and use HPLC-ELSD to determinate the content of kinds of amino acids in ginseng. To analysis the content tends of total amino acids, essential amino acids, and finally we proceeded the system cluster analysis on acids.Use 80% ethanol to extract and HPLC method to detect the contents of 7 kinds of organic acids in the ginseng, and analysis its content trends of ginseng in different years and origins, and finally we carried on the system clustering analysis on it.Use ultrasonic oscillations instrument to extract and HPLC method to detect the content of nucleoside compounds in ginseng, and analysis its content trends of ginseng in different years and origins, and we carried on the system clustering analysis on it. Finally we established the HPLC fingerprint of nucleoside compounds.
