Our aim was to discover pivotal dysregulated proteins which are biomarkers in the multiple drug resistance (MDR) of human hepatocellular carcinoma (HCC). To do so we comparatively analyzed the secretome profiles of human HCC MDR cell line BEL7402/5-FU and its parental cell line BEL7402 using iTRAQ-coupled LC-MS/MS. In total, we identified 229 differentially expressed proteins, of which 131 proteins were over-expressed in BEL7402/5-FU related to its parental cell line, and 98 proteins were under-expressed. We further validated several differentially expressed proteins by real-time PCR and western blot analysis. We also determined the association of MDR with one of the highly regulated proteins, AHSG This study details the application of iTRAQ technology to MDR biomarkers in HCC cell secretome. The differentially expressed proteins which may be linked to MDR of HCC represent valuable additional information in our understanding of MDR.
