Background:Tumor cells are resistant to chemotherapeutic drugs. Drug resistance was correlated with tumor stem cells. How to kill tumor stem cells will become the key to the treatment of oral squamous cell carcinoma. Objective:to study on the effect of 5-fluorouracil on Tca8113 cells, we determined cell activity、cell cycle and the related marker of tumor stem cells of Tca8113 cells. Method:with the different concentrations of 5-fluorouracil and the different duration of drug action, Tca8113 cells were determined by CCK-8 to understand the effect of 5-fluorouracil on cell activity and to try to find the best drug concentration and time. Then the cell cycle and the side cell in Tca8113 cells were determined by flow cytometry when they are added with 5-fluorouracil. In the end, we used the method of scratch test to determine Tca8113 cells migration before and after using 5-fluorouracil. Result and conclusion: different concentration and time of 5-fluorouracil using for enriching tumor stem cells were determined as 50μg/ml with 48h incubation. After enrichment, G0/G1 phase cells increase from 51.06%±0.74% to 73.12%±3.68%(P=0.01). At the same condition, S phase cells decreased from 30.53%±0.33% to 26.88%±3.68%(P=0.244), G2/M phase cells were disappeared in experimental group (using 5-fluorouracil), and control group was 18.42%±0.47%(P=0.00). In summary, the proportion of the cell cycle in Tca8113 cells has significant statistical differences. The proportion of side population cell (SP cell) which can be stained by heochest33342 in Tca8113 cells experimental group increased from 0.24%±0.02% to 0.52%±0.02%, the difference was also significant (P=0.00). The scratch test showed that the experimental group cells’ability of wound healing was greater than the control group’obviously.5-fluorouracil can enrich the cancer stem cell population in cell line Tca8113 cells. Drug resistance may be related with tumor stem cells.
