Objective To investigate the effects of leptin on the differentiation of RAW264.7 cells to osteoclasts induced by soluble receptor activator of nuclear factor-κ B ligand (sRANKL) and its possible mechanisms. Methods The effects of leptin on the differentiation from osteoclast precursor cells to osteoclasts were detected by tartrate-resistant acid phosphatase(TRAP) staining. The mRNA expressions of TRAP、CTSK、 c-jun、c-fos and peroxisome proliferator-activated receptory (PPARy) in osteoclast precursor cells were measured by quantitative real-time PCR(qRT-PCR). The protein expressions of TRAP、CTSK、c-jun、c-fos and PPARywere detected by western blot. Results Compared with sRANKL treatment group, the differentiation from osteoclast precursor cells to osteoclasts was significantly inhibited in sRANKL combination with leptin treatment groups. Additionally, the mRNA and protein expressions of TRAP,CTSK,c-fos and PPARywere significantly decreased in sRANKL combination with leptin treatment groups compared with the sRANKL treatment alone (all P<0.05 or P<0.01). Furthermore, the expression of PPARy was decreased in a dose-dependent manner(P<0.01). However,the mRNA and protein expressions of c-jun were significantly increased in sRANKL combination with leptin treatment groups compared with the sRANKL treatment alone (all P<0.05 or P<0.01).Conclusion Leptin could inhibit the differentiation from RAW264.7 cells to osteoclasts via inhibiting the expression of PPARy.
