Objective:The purpose of this experiment was to investigate the expression of Beclin 1、 GLUT-1 and HIF-1α in head and neck squamous, and the relationship between they and the clinical stage, tumor location, histological grade and lymph node metastasis.Materials and methods:Use the immune tissue test to detect Beclin 1、GLUT-1 and HIF-la in 29 cases with head and neck squamous cell carcinoma and 10 cases of adjacent tissues, including 12 cases of laryngeal cancer,17 cases hypopharyngeal. All cases They are confirmed by operation or biopsy.Result:1. General Information:29 cases of head and neck squamous cell carcinoma, all male, aged 33 to 79 years,60.4 years old in anverage. Among 12 cases (41.4%) of laryngeal cancer, including 10 cases in the supraglottic area,2 in the supraglottic region; 17 cases (58.6%) of hypopharyngeal cancer, including 15 cases of pyriform type,1 case of posterior pharyngeal wall type, one case after ring type.26 patients of them (89.6%) had a history of smoking,21 patients of them (72.4%) were addicted to drink, and 19 patients of them (65.5%) addicted to both smoking and drinking. TNM classification:4 cases T1N0M0,3 cases T2N0M0,1 cases T1N2M0,6 cases T2N1M0,2 case T2N2M0, 3 case T3N2M0,3 case T3N1M0,3case T4N1M0,4 cases T4N2M0; clinical stages4:Ⅰ stage (13.8%), II stage (10.3%), III stage (31.0%), IV stage (44.8%).2. Beclin-1:the average number of optical density in para carcinoma tissue is 0.0345±0.0036, in head and neck squamous cell carcinoma is 0.0107±0.0017, which is lower than in para carcinoma tissue.There was significant difference (p<0.001) between them. Beclin-1 in laryngeal carcinoma and adjacent laryngeal carcinoma t optical density in the average were 0.0117±0.0035,0.0397±0.0046, Beclin-1 expression in laryngeal carcinoma is lower than adjacent laryngeal carcinoma (p<0.001); Beclin-1 in the hypopharynx carcinoma, and adjacent hypopharyngeal carcinoma optical density in the average were 0.0099±0.0016,0.0292±0.0048, Beclin-1 expression in hypopharyngeal tissues is lower than in adjacent hypopharyngeal carcinoma (p<0.001).3. Expression of GLUT-1:the average number of optical density in adjacent tissues of GLUT-1 is 0.0147±0.0023, the average number of GLUT-1 in head and neck squamous cell of optical density is 0.0273±0.0034, GLUT-1 expression in head and neck squamous cell carcinoma is higher than in adjacent tissues, but there are significant difference (p=0.044). The averages of GLUT-1 in laryngeal carcinoma,and laryngeal carcinoma adjacent optical density is 0.0298±0.0047,0.0162±0.0018, GLUT-1 expression in laryngeal tissue is higher than in adjacent laryngeal carcinoma, but there are no significant difference (p=0.258). The averages of GLUT-1 in hypopharyngeal tissue adjacent optical density and hypopharyngeal carcinoma were 0.0298±0.0047, 0.0162±0.0018, GLUT-1 expression in hypopharyngeal cancer is higher than adjacent tissue, but there are no significant difference (p=0.086).4. The expression of HIF-la:The averages of adjacent tissues HIF-la optical density was 0.0120±0.0012, The averages of head and neck squamous cell carcinoma tissue HIF-la optical density is 0.0271±0.0025, HIF-1a of head and neck squamous cell carcinoma expressionis higher than adjacent tissues there was significant difference (p=0.001). Stratified, the average optical density of HIF-la in laryngeal carcinoma and adjacent laryngeal carcinoma are 0.0314±0.0046,0.0123±0.0024, The average optical density of HIF-la in laryngeal carcinoma higher than the adjacent laryngeal carcinoma, but there are no significant difference (p=0.068); the average optical density of HIF-la expression in hypopharynx carcinoma, and adjacent hypopharyngeal carcinoma are 0.0254±0.0031,0.0119±0.0014, the average optical density of HIF-la in hypopharynx carcinoma is higher than adjacen hypopharyngeal carcinoma, a significant difference (p =0.014).5. In the expression of Beclin-1 of clinicopathological factors in 29 cases of head and neck squamous cell carcinoma,there are no significant relationship with age, lymph node metastasis, T stage, clinical stage, location, smoking, drinking and there are no significant difference.6. The relationship between GLUT-1 and age:THE expression of GLUT-1 in the higher age group (≥60 years) (0.0323±0.0044) higher than lower age group (<60 years) (0.0164±0.0039), the difference was significant (t=2.47, p=0.02); THE expression of GLUT-1 in the late expression of lymph node metastasis of head and neck squamous cell is higher than earlier carcinoma no expression of lymph node metastasis of head and neck squamous cell carcinoma, but no significant difference, There are no differences. with location, smoking, drinking.7. In the expression of HIF-1α of clinicopathological factors in 29 cases of head and neck squamous cell carcinoma, there are no significant relationship with age, lymph node metastasis, T stage, clinical stage, location, smoking, drinking and there are no significant difference.Conclusion:So the occurrence and development maybe related to head and neck squamous cell carcinoma are related to the abnormal expression of Beclin 1、GLUT-1 and HIF-1α.
