This research is supported by the Natural Science Foundation of China——The research of process mechanism that Jiangyou Aconite(Heishunpian) was concocted in the original place to reach "the attenuated deposit efficiency" of "four rules and three principles" (grant No.81274086).Fuzi, as one of the toxicant traditional Chinese medicines, is the processed lateral root of Aconitum carmichaelii Debx, and needs to be processed before clinical useage generally. Commercial specification of Fuzi mainly includes Heishunpian, Baifupian, Danfupian, Paofuzi and Yanfuzi, of which Heishunpian has the largest circulation. The detailed process procedure is as follows:It must be cleaned up and soaked in mineral salt solution no less than 7 days to avoid decay, then put it out and boil with clean water about 30 minutes. After it is thoroughly boiled, it is need to be immersed with clean water for 1 night, the aim of this procedure was to cool it down as quickly as possible. Take it out and cut into 0.5 cm longitudinal slices the following day, rinsed with water again and again to get the product which has no mineral salt. After rinsing, it must be continued steaming for at least 6 hours. Finally, it is need to be baked to well-dryness to get Heishunpian. About it’s quality evaluation, modern research was usually focused on the raw and processed products in components, efficacy, and toxicity. Given these, this paper will study the composition change through traditional analytical technologies, in combined with intelligent sensory technologies to explore the "taste" change of the process, and aim to clarify the process mechanism.Firstly, this research teases out process link points of Heishunpian through on-the-spot investigation, and provides a basis for follow-up study.Secondly, this paper was mainly focused on danba, polysaccharides, total alkaloids, and 6 index components in contents analysis, and study the characteristic fingerprints variation of alkloids fraction of the process of Heishunpian, and in combined with HPLC-MS to analyze the unknown components qualitatively The results showed that the contents of danba increased in "paodanv link, and thereafter decreased along with the process. Polysaccharides and total alkaloids had a decreasing tendency. Different treatments had different effect on the index components of Fuzi, it’s more obvious by heating treatment. As the process continues, the contents of diester diterpene alkaloids were decreased step by step, which is in accordance with its processing mechanism, while the contents variation of monoester diterpene alkaloids were not obvious.Furthermore, to preliminarily explore ’taste" of the process of Heishunpian through electronic tongue, variation tendency was clarified. The results showed "taste" variation is closely related to the introduction of danba in the process of Heishunpian.In order to explore whether electronic nose is useful for identification of yinpian whose favor is micro, heishunpian was choosed to study.20 batches of Heishunpian were collected, which included different producing areas and grades. Using electronic nose technology to test, then obtain the sponse values from the sensors, and the data was processed through principal component analysis (PCA), discriminant factor analysis (DFA). The results showed that electronic nose can be used for differentiation of Heishunpian.In conclusion, this research applies traditional analytical technolygies and electronic tongue to study variation of contents and "taste", and to explore the possiblitity to distinguish yinpian by electronic nose. This paper revealed that change rule of main contents of the process of Heishunpian, and clarify that "taste" variation is correlated with the introduction of danba preliminarily, and also proved that electronic nose can be used for distinguishing the producing areas and grades of Heishunpian, of which "favor" is not obvious. This topic is a good help to broaden processing ideas for other traditional Chinese medincines.
