Objective:the aim of this study was to investigate the time phase characteristics of locomotor activity rhythms of circadian-disorder mice, and the effect of electro-acupuncture (EA) on their locomotor activity rhythms and to explore the liver circadian related genes involved in the mechanism of action of adjusting rhythm disorders.Methods:The circadian rhythms of locomotor activity of the circadian-disorder mice were monitored and recorded by Data Acquisition and Processing System for locomotor activity rhythm for Small Animals made by Chengdu University of TCM and ClockLab Running Wheel Data Acquisition System for Circadian Biology made in USA.24 animals which were screened out were divided randomly into 4 groups which were blank group,model group,electro-acupuncture and natural recovery group.Each group received LD (Light:06:00-18:00, Dark:18:00-06:00) domestication for 10 days. After domestication, every 2 days will turn on the light time is 8 hours advanced for 10 days totally and restore natural light(Light: 06:00-18:00, Dark:18:00-06:00).After that, the blank group and the model group was executed and the electro-acupuncture group was administered on electro-acupuncture treatment (electro-acupuncture was administered at Ganshu(BL18) and Zhiyang(GV9), on ZT16 with HANS Acupoint and Nerves Stimulator (HANS-200) using 2/15Hz and 0.5mA for 15 min each time and one time a day).The natural recovery group not curative,but bundled for balance stimulus.After the third electro-acupuncture, put to death animals materials under test.Use the real-time fluorescence quatititive PCR-Array to test the change of the liver circadian related genes.Result:(1)Before model eastablishment, there were no statistical difference in spontaneous activity cycle, acrophase of circadian rhythm, total activities and onset time point in the blank group and the model group anmals (P>0.05.After model eastablishment,there were statistically significant between the model group and the blank group, show that spontaneous activity cycle, acrophase of circadian rhythm, total activities and onset time point(P<0.05).(2)After electro-acupuncture,there were statistically significant in acrophase of circadian rhythm, onset point and total activities of spontaneous activity in electric acupuncture group (P<0.05);There were statistically significant in acrophase of circadian rhythm of spontaneous activity in natural recovery group mice (P< 0.05);There were no statistically significant in period of spontaneous activities between electric acupuncture group and the natural recovery group (P> 0.05) (3)The model group compared with the blank group,the expression of circadian clock gene Bmall, Fbxl3 and melatonin inhibitors Mtnrla rise, while the expression of the circadian clock gene DEC2 decline;The electro-acupuncture group compared with the model group, the expression of circadian clock gene Bmall, Fbxl3 and melatonin inhibitors Mtnrla decline,while the expression of the circadian clock gene DEC2 rise;The natural recovery group compared with the model group, the expression of circadian clock gene Bmall,DEC2, Fbxl3 and melatonin inhibitors Mtnrla unchanged.Conclusion:After model eastablishment, the spontaneous activity circadian rhythm have changed, and the phase characteristics was the period cycle shortened, the acrophase and onset point advanced, and total activites declined;The electro-acupuncture can restore the arcophase and onset point in time,, but not significantly affect the period and the total activity decreased;The up regulated of electro-acupuncture on chronic jet-lag gene Bmall, Fbxl3 and melatonin inhibitors Mtnrla and the down regulated effect of electro-acupuncture on DEC2 may be one of molecular mechanism of electro-acupuncture regulated the circadian.
