Objective In the foundation of early filtering optimization and getting a better compatibility party of Rhizoma Curculiginis and Epimedium enhancing efficacy and reducing toxicity, further study the influence of compatibility and single party optimized on invigorantion of the kidney yang deficiency mice, and analyze and discuss the physiological mechanisms of compatibility of ’Drug Douple’Rhizoma Curculiginis and Epimedium may improve the kidney yang deficiency symptoms.Methods According to the weight value,77 ICR mice were randomly divided into 7 groups, namely normal group, model group, positive group, Curculigo-Epimedium 1:2 group, Curculigo-Epimedium 1:1 group, Curculigo-Epimedium 1:0 group, Curculigo-Epimedium 0:1 group. In addition to the normal group, the other groups of mice were fed with adenine suspension daily. After one hour, each treatment group given corresponding medicines, and the normal group and model group were given the same amount of distilled water for 4 weeks. Detect body weight, food intake and water intake of mice in each group every week. Mice were detected temperature after drug for 2 weeks and 4weeks. In the end of the experiment, weight liver, bilateral kidneys, thymus, testes tissue, and calculate viscera index. Detect the serum ACTH and liver CYP activity. By fluorescence quantitative RT-PCR and Western Blot method, the expression of each group of mice liver CYP1A2, CYP2E1, CYP3A11 and kidney OAT-1, OAT-3, OCT-2 mRNA and protein were measured.Results Compared with the normal group, model group have a significant decreasing on body weight, food intake, body temperature, organ (liver, kidney, thymus, testis) weight, organ index (thymus), serum levels of ACTH, enzymes (CYP1A2, CYP3A11) activity, protein (CYP1A2, CYP3A11, OCT2) expression levels and other indicators, while have a significant increasing on the enzyme (CYP2E1) and protein (CYP2E1, OAT1, OAT3) expression. Compared with the model group, the compatibility party have a significant rising on body weight, organs (liver, kidney, testes) weights and organ (thymus) index, body temperature, serum levels of ACTH, enzymes (CYP1A2, CYP2E1) activity, protein (CYP1A2, OCT2) expression levels and other indicators, and have a significant declining on the enzyme (CYP2E1) activity, protein (CYP2E1,OAT1,OAT3) expression levels and other indicators. single party group have a significant rising on organs (liver, kidney, testis) weight and dirty device (thymus) index, protein (CYP1A2, CYP3A11, OCT2) expression levels and other indicators, while have a significant declining on the protein (CYP2E1,OAT1,OAT3) expression levels and other indicators. Compared with the single group, the compatibility party have a significant increasing on body temperature, liver weight, protein (CYP1A2, OCT2) expression, while have a significant declining on protein (OAT1,OAT3, CYP2E1) expression levels.Conclusion The compatibility party have a better therapeutic effect than single party, its best compatibility proportion is Curculigo-Epimedium 1:2 and its mechanisms to improve the symptoms of deficiency may be adjusted by the following two conditions common:①by adjusting the adrenal steroid hormones and enzymes (liver CYP1A2, CYP2E1);②by down-regulating the expression of renal ion transporters OAT1, OAT3 and up-regulating OCT2, reduce the accumulation of anionic species in renal tubular epithelial cells, which play a protective effect on the kidneys, while results show that hepatic metabolism enzyme CYP1A2 and CYP2E1 may exist antagonistic pharmacology and toxicology, Compatibility of ’Drug Couple’Rhizoma Curculiginis and Epimedium may influence the dynamic balance between the two enzymes so as to improve the kidney yang deficiency symptoms in mice.
