Objective: Ubiquitin specific protease 7(USP7) has important contact with the tumor in occurrence and development, the existing research has shown that USP7 is highly expressed in laryngeal cancer tissues, and the laryngeal cancer’s cell proliferation, cell cycle and cell invasion ability obviously changed after knocked down the USP7, but the mechanism is unknown.This research through the siRNA(small interfering RNA) konck down the USP7 expression in laryngeal cancer Hep2 cells, through the transcriptome sequencing screening the differentially expressed genes, to explore the mechanism of USP7 on the biological behavior of laryngeal cancer cells.Methods: Design specific siRNA to transfect laryngeal cancer Hep2 cells by virus and specificly konck down the expression of USP7, after extracting mRNA through the transcriptome sequencing screen and analyse the differentially expressed genes of knocked down USP7 in laryngeal cancer cells.Results:1.The specific siRNA significantly knocked down the USP7 expression in laryngeal cancer cells.2.Through the transcriptome sequencing technology, compared with the laryngeal cancer cell group which did not give any treatment,we found that the laryngeal cancer cells knocked down the USP7 have 57 genes raised(60.6%), 37 genes lowered(39.4%).And after expanding the differences multiples(4 times or higher), found SULT1A4, MTPN, CTF2IP1, BUSP5, SCLY, PI3 etc 22 significantly raised, SULT1A3, TICAM2, MUC13, ARL17 B etc 11 genes significantly lowered.3.Through the GO function enrichment analysis found that the differences in genes mainly concentrated in the cell cycle and proliferation, metabolism, positive or negative regulation and control, enzyme catalytic activity, apoptosis, signal transduction, immune response, etc.Conclusion: USP7 to laryngeal cancer cell’s proliferation, cell cycle and cell invasive ability etc biological behavior influence mechanisms possiblely involved on various aspects, mainly related with the cell cycle and proliferation, metabolism, positive or negative regulation and control, enzyme catalytic activity, apoptosis, signal transduction, immune response etc, some of the differentially expressed genes could be the potential targets for the biology treatment of laryngeal cancer cell, but it is necessary for further verification.
