In this study, compounds(DFG) were extracted from the corn bran with dilute alkali and enzyme, which contain 74.2% of polysaccharides, 6.78% of phenolic and 16.8% of proteins. The average molecular weight is 17.9kDa.We investigate the antioxidant activity of DFG. DFG could scavenge free radicals to 94.13% when the concentration was 12 μM measured by the DPPH method. When determined by FRAP method, the total antioxidant capacity of DFG was better than positive control and it showed obvious effect at low concentration. DFG restored the oxidative damage in B16 melanoma cells in a dose-dependent manner. The restore rate is 92.9% when the concentration was 0.3μM.We investigate the anti-aging capacity and the mechanism of DFG by using caenorhabditis elegans(C.elegans) as model. DFG eliminated 50.9% of the ROS in C.elegans, and extended the lifespan to 22% and 16.7% respectively under the heat shock and natural conditions when the concentration was 1.5mg/m L. Further studies showed that this effect was related to the daf-16 and sir-2.1 factors in the nematode.DFG could make the DAF-16 enter the nucleus observed by fluorescence confocal microscopy. DFG could up-regulated the expression of SOD-3 factor to 20% determined by fluorescence microplate reader. The results showed that DFG can play a role in anti-aging by stimulating the DAF-16 factor into the nucleus and enhancing its transcriptional activity.DFG could up-regulate the expression of sir-2.1 factor to 5.2 fold. And when simultaneous administrated with an SIR-2.1 factor enzyme activity inhibitor(NAM) the anti-aging capacity of DFG was lost. The results show that DFG could extend lifespan by up-regulating the expression of sir-2.1 factor and depending on the acetyl enzyme activity of SIR-2.1.
