The active polypeptide extracted from frog skin, which contains many amio acids and cytokines, It has been reported to promote skin repair and regeneration. and has a wide range of biological effects, it can be widely used in medicine.But Rana leather polypeptide whether can inhibit apoptosis, or promote wound repair situation is not yet explained.This study intends to apply purified Rana polypeptide gel formulation to the dorsal skin of rabbits trauma test, explore the wound repairing of frog skin peptide.Firstly, 30 m J/cm2 skin exposure to ultraviolet radiation induced apoptosis established by simulating solar ultraviolet; assess whether frog skin peptide can inhibite the apoptosis of Ha Ca T cells. Secondly, regard the full-thickness skin injury in New Zealand white rabbits as model for assessment of wound healing, HE staining, calculate the rate of wound healing. Finally, a compr ehensive analysis of frog skin peptides on wound healing.The results in Ha Ca T cells of apoptosis with frog skin polypeptide show that: compared with the control group, the apoptosis rate significantly increased under the condition of UVB, under conditions of UVB, after the EPR800, APR800, the increased apoptosis rate of Ha Ca T cells is significantly inhibited; In the wound macroscopic evaluation, we found that the frog skin peptide in the experimental group got slightly lower scores in lammatory swelling,and shorter time in crusting, which compared with the control group. And frog skin polypeptide experimental group dose group(20mg / m L) has the optimal healing effect; The results of healing rate showed that: 1,3,6,9,12 day, frog skin peptide C1, C2, C3 compared with the negative control group, the healing rate was increased significantly; wherein, the healing rate effect of C2 group is more significant; The results of Histopathological changes showed that: The numbers of inflammatory cells infiltration in frog skin polypeptide experimental group was significantly decreased, compared with negative control groups. In the C2 and C3 experimental group,the numbers of capillaries and fibroblasts of nascent hyperplasia are significantly more than the negative control group, which is conceived to the proliferation of wound repair.In summary, the frog skin peptide can inhibit apoptosis of Ha Ca T cells; Frog skin peptide can promote the repair of full-thickness skin wounds in rabbits; Among the experimental group frog skin polypeptide dose group(20mg/m L) concentration group has the best wound healing effect,which has the shortert healing time, scab fastest.
