Analysis Of Association Between Uncontrolled Asthma Patients And Glucocorticoid Receptor Gene Polymorphism

Objective:To investigate the correlation between Glucocorticoid receptor rs4912911、rs6191 and rs6198 gene polymorphisms and refractory asthma. Method:Dec 2014 to Nov 2014, in our hospital diagnosed as bronchial asthma patient and inhaled corticosteroids therapy for half a year, divided into three group:30 cases of no control group, 30 cases of control group, 30 cases of healthy control group,All subject of the three groups were genotyped by Sequenom MassARRAY ? SNP assay and matrix-assisted laser desorption/ionization time of flight(MALDI-TOF)method for three polymorphisms. Result: rs4912911-GG/GA/AA genotype frequency respectively in three groups: no control group 16.7%, 33.3%, 46.7%, control group 10%, 53.3%, 36.7%,healthy control group 23.3%, 53.3%, 23.3%; rs6191-GG/GA/AA genotype frequency respectively in three groups: no control group 55.2%, 55.2%, 37.9%, control group 50%,46.7%, 3.3%, and the healthy control group were 36.7%, 60%, 3.3%, accord with Hardy-Weinberg genetic balance, comparing two differences groups have no statistical significance(P < 0.05); rs6198-AA/GA/GG genotype frequency respectively in three groups: no control group 30%, 33.3%, 36.7%, control group 46.7%, 43.3%, 10%, healthy control group 60%, 33.3%, 6.7%; compared with no control groups and control groups was statistically significant(2? = 6.05, P = 0.049, P < 0.05), GG genotype significantly higher in no control group, GG genotype may be associated with asthma control situation.Conclusion: rs4912911, rs6191, rs6198 gene exist polymorphism, this study found rs4912911 and rs6191 gene polymorphism no related with control situation in bronchial asthma, rs6198 gene polymorphism may be related to refractory asthma, G allele gene may be susceptibility genes of steroid resistance.
