In this paper, the extraction and purification technology of ginger essential oil (GEO) was studied, and the optimum technological conditions were obtained. Established the animal skin inflammation model, studied the anti-inflammatory activity of GEO and the effect of GEO on allergic contact dermatitis (ACD) at molecular level. The main research contents and results are as follows:1. The extraction of GEO. Supercritical carbon dioxide extraction was used to extract GEO, and the optimal extraction conditions were obtained by single factor test and response surface analysis. The conditions are as follows:extraction temperature 40.85 ℃, extraction pressure 22.59MPa, extraction time 4.23h, and the yield of GEO in this condition 1.2314%. The actual operation will be adjusted to:extraction temperature 41℃, extraction pressure 22.6MPa, extraction time 4.23h, the actual yield of GEO 1.23 ±0.0043% and the purity 16.01±0.31%.2. The purification of GEO. The dynamic adsorption and desorption experiments of AB-8 macroporous resin for extraction of GEO from D101, AB-8, D1300 and DA201 were carried out by static adsorption test. Study the sample concentration, sample flow rate, ratio of ethanol/water and elute flow rate. And the optimal conditions of macroporous resin purification are as follows:the sample concentration 3mg/mL, the flow rate 3mL/min,100% ethanol as the eluent and the elute flow rate 0.5mL/min. The purity of GEO obtained under this condition was increased from 16.01% to 46.60%. The results showed that AB-8 macroporous resin had good purification effect on GEO.3. The anti-inflammatory activity of GEO. Xylene induced ear swelling of mice, carrageenin induced paw edema of mice, xylene induced mouse skin capillary permeability changes, and allergic contact dermatitis (ACD) test were used to evaluate the activity of GEO of anti-inflammatory activity. The test results showed that GEO can reduce the mice ear and paw swelling, inhibit skin capillary permeability. In the test of ACD, GEO can significantly inhibit the swelling rate and improve the skin lesions. Histopathological observations indicated that GEO can reduce inflammatory cell infiltration, cell swelling and vasodilatation. They were showed that GEO on acute skin inflammation and chronic allergic inflammation contact dermatitis has good effect and has obvious dermatitis inhibition.4. The anti-inflammatory mechanism of GEO on ACD. In the course of ACD, many cytokines play an important role. In this paper, we study the toll-like receptor 2 (TLR-2), toll-like receptor 4 (TLR-4), tumor necrosis factor alpha (TNF-α), interferon gamma (IFN-γ), interleukin 1β (IL-1β), interleukin 8 (IL-8) and interleukin 4 (IL-4) seven kinds of cytokines in ACD. RT-PCR was used to research and it was found that GEO can cut mice ear tissue and spleen in TLR-2, TLR-4, TNF-α, IFN-γ, IL-1β and IL-8 gene expression, increased the gene expression of IL-4. The mechanism of the anti dermatitis of GEO may be related to the regulation of the expression of inflammatory cytokines.In summary, using supercritical carbon dioxide extraction combined with AB-8 macroporous resin can achieve good extraction and purification effect of GEO. GEO has obvious anti-inflammatory activity, and its anti ACD mechanism is related to the regulation of gene expression of inflammatory cytokines.
