Objective To investigate the changes of IL-6/STAT3 signaling pathway in subtotal hepatectomy after Echinococcus multilocularis(E.m.) infection and the effective to liver regeneration. Methods Forty C57 mice were infected with E.m. suspension through subcutaneous injection, and feed forty normal C57 mice as control group. Then eight mice was chosen randomly as baseline in each group, those mice were so called non-operation. Three months after the inoculation, those mice were taken 2/3 hepatectomy in each group, and sacrificed at the 1st, 2nd, 4th, 7th day after operation, then took liver and peripheral blood for the analysis. peripheral blood was taken to test the IL-6 level by ELISA, and liver tissue was taken to test the expression of IL-6 m RNA and STAT3 protein by Taqman Real-time PCR and Western Blot. Then analysis the results. Result The peripheral blood’s ALT, AST level at 1st day after 2/3 hepatectomy were increasing obviously, then declined to normal gradually. And there were no significant difference between two groups in each days after operation. The peripheral blood’s IL-6 expression in the base group and 1st, 2nd, 4th, 7th day after 2/3 hepatectomy in the experiental and control groups, respectively(6.31±0.366,5.74±0.053),(6.14±0.146, 8.30±1.799),(5.93±0.144,6.36±1.148),(5.77±0.064,5.88±0.089),(6.02±0.400,5.93±0.042), and there were significant difference in the base and 1st day after operation(Ubase = 7.000, Pbase = 0.025),(U1 = 3.000, P1= 0.004).While the other groups were no statistically significant difference. the IL-6 m RNA expression of liver tissue in the base group and 1st, 2nd, 4th, 7th day after 2/3 hepatectomy of experimental and control groups were respectively(13.33±2.133, 14.08±0.914),(12.86±2.661, 13.30±9.093),(15.246±3.163, 15.867±0.907),(15.608±0.977,11.719±2.335),(14.004±2.567, 16.079±0.452), and there were significant difference in the 4th and 7th day after operation(U4 = 0.000, P4 = 0.002),(U7 = 5.000, P7 = 0.013), and othergroups were no statistically significant difference. The level of STAT3 protein of liver tissue in the base group and 1st, 2nd, 4th, 7th day after 2/3 hepatectomy of the experimental and the control groups are respectively(0.497±0.002, 0.558±0.000),(0.271±0.007, 0.340±0.006),(0.127±0.012, 0.212±0.072),(0.183±0.018, 0.056±0.015),(0.210±0.037, 0.116±0.040), and there were significant difference in the 2nd, 4th and 7th day after operation(U2 = 0.000, P2 = 0.004),(U4 = 0.000, P4 = 0.004),(U7 = 2.000, P7 = 0.010) and other groups were no statistically significant difference. The Brd U expression in liver tissue in the base group and 1st, 2nd, 4th, 7th day after 2/3 hepatectomy of the experimental and the control groups, respectively(1.67±0.745 、 2.33±0.943),(9.17±1.344、22.50±2.500),(14.33±1.491、8.17±1.572),(3.67±0.933、2.33±0.943), and there were significant difference in the 2nd and 4th day after operatio(U2=0.000,P2=0.004),(U4=0.000,P4=0.004) and the other groups were no statistically significant difference. And there were no obvious expression of CD31 in liver tissue in the two groups. Conclusion E.m. infection may cause the delay function to the launch and time for taking the maximum capacity of liver regeneration after 2/3 hepatectomy.
