Urumqi Uygur And Han Patients With Asthma Of Peripheral Blood IL-2, IL-4, IFN-γ Gene Expression And Methylation Levels Of Specific Research

Objective:This study aims to detect the mRNA expressions of interleukin IL-2, IL-4 and IFN-γ of CD4+T lymphocyte as well as the protein expression levels of plasma IL-2, IL-4 and IFN-γ in peripheral blood of asthma patients and healthy populations from the Uyghur and Han nationality, and also to detect the methylation levels of IL-4 and IFN-γ, and to compare the correlation of gene expression and methylation levels. Methods(1) Study subjects were 150 patients with acute exacerbation of asthma(asthma group) and 107 healthy people(control group). The asthma patients included 30 Uygur and 120 Han people, while the controls included 35 Uygur and 72 Han people. People of the case group were diagnosed with acute episode of asthma at the First Affiliated Hospital of Xinjiang Medical University and the Xinjiang Uygur Autonomous Region people’s Hospital I,while people of the control group were the health examination people who carried out physical check-up in the First Affiliated Hospital of Xinjiang Medical University during the same period.(2)Adopt the SYBR Green I real-time quantitative RT-PCR detect IFN-γ,IL-2 and IL-4 mRNA gene expression level.(3)Adopt ELISA detect the IFN-γ,IL-2 and IL-4 protein expression level.(4)Adopt the SNaPshot to have the methylated quatitation analysis on the 107 samples of asthma group and 105 samples of IL-4,also 5 adjacent CpG sites was analyzed by single base extension of IFN-gamma.Results:(1) The expression levels of IL-2mRNA and protein in asthma group were lower than control group, and the difference has no statistically significant(P>0.05),there is no statistically significant difference between Uygur and Han asthma patients(P>0.05).The expression of IL-4,IFN-γmRNA and protein in asthma group were higher than control group, the difference was statistically significant(P<0.05),the IFN-γ mRNA and protein expression level in Uygur and Han asthma patients had significant difference(P<0.05),the IL-4mRNA expression in Uygur asthma and Han asthma had significant difference(P<0.05),IL-4 protein expression in Uygur asthma and Han asthma had no significant difference(P>0.05).(2)IFN-γ1SF,IFN-γ- 2SR,IL-42 SR,IL-44 SR methylation degree had no statistically signification difference on asthma and control group(P>0.05).IL4-1SF methylate degree had statistatically signification differen-ce on asthma and control group(P<0.05). The degree of methylation of IL-41 SF locus in Han asthma group was higher than that in the Han control group, the difference was statistically significant(P<0.05).IFN-γ1SF,IFN-γ-2SR,IL-41 SF,IL-42 SR,IL-44 SR methylate degree had no signification difference on Uighur asthma and Han asthma group(3)IFN-γ,IL-4mRNA expression level and IFN-γ1SF,IFN-γ-2SR,IL-41 SF, IL-42 SR, IL-44-SR methylate degree has no correlation(P>0.05).Conclusion:The expression of IL-4 and IFN-γmRNAand protein have statistically significant in asthma group and control group.The IL-4mRNA expression in Uygur asthma and Han asthma differences were statistically significant, the IL-4 protein expression in Uygur asthma and Han asthma had no difference.The IL-2mRNA and protein expression levels in asthma group and the control group were not statistically significant difference, and the expression of mRNA in the Uygur asthma patients and Han asthma patients have no obvious difference,while the protein expression difference has statistical significance.IFN-γ has statistatically signification difference on the gene expression and protein expression difference in Uygur and Han asthma group,may the reason of the prevalence difference in Uygur and Han asthma group.IL4-1SF has difference between the asthma and control group.IL-41 SF site methy-lation can be the way to study asthma.There was a significant difference between the IL4-1SF locus in the Han control group and Han asthma group,there was no difference between the Uygur asthmatic group and the Uygur control group, it is suggested that this locus is more likely to occur in Han population in hypermethylated.The asthma group IL-4,IFN-γ methylation levels in the promoter region of degree has no regulation function on mRNA level.IL-4,IFN-γ has no difference with Uygur and Han asthma group,proved that IL-4 and IFN-γ DNA methylation may not be the cause of the different prevalence of Uygur and Han asthma.
