Objective:Lymph node metastasis of gastric cancer has been a difficult problem, and the animal model is an important method to study the lymph node metastasis of gastric cancer. At present, there is still a lack of a classical animal model to simulate the process of lymph node metastasis, and the selection of experimental animals is also lack of standards. The aim of this study was to screen the experimental animals and tumor inoculation methods appropriate to the study of lymph node metastasis of gastric cancer, and to lay the foundation for the establishment of the animal model of lymph node metastasis in gastric cancer.Methods:一.The feasibility of the model of gastric cancer in nude mice and New Zealand white rabbits1 6 BALB/C-nu/nu nude mice and New Zealand rabbits were injected with HGC27 cells in the gastric mucous respectively;2 6 New Zealand white rabbits were injected with tissue suspension VX2 in gastric mucous respectively;3 compare the results of gastric cancer which results from HGC27 and suspension VX2 respectively.二. The characteristics of two different methods for the in situ inoculation of cell suspension and tumor tissue mass were compared.65 BALB/C-nu/nu nude mice were randomly divided into two groups,30 in group A and 35 in group B;In group A, mice were injected with human gastric cancer HGC27 cell suspension into the gastric wall;In group B firstly five nude mice were injected hgc27 cell suspension liquid subcutaneously, Remove the tumor piece until the size of nude mice subcutaneous tumor blocks reaching 1 cn* 1cm, put the blocks into RPMI-1640 and cut them into tubercles of about 1 mm3, tubercles were in situ inoculated to remaining 30 nude mice stomach;2.Take lymph nodes from gastric and surrounding tissues together with mesenteric lymph nodes and make pathological examination of liver, spleen, kidney and mesenteric lymph nodes in 28 days,21 days and 14 days respectively;3.compare the results of 3 groups in gastric cancer formation, peritoneal metastasis, ascites formation and lymph node metastasis.Results:1.Application of human gastric cancer HGC27 cell line in nude mice succeed establishing the model of lymphatic metastasis of gastric cancer while in New Zealand white rabbits failed;2.VX2 tissue block can induce lymph node metastasis model in New Zealand white rabbits, however because VX2 is a rabbit derived epithelial squamous cell carcinoma, there are some differences between human gastric cancer cell line in the biological characteristics.3. In group A the gastric cancer model was established using HGC27 cell suspension. The gastric in situ tumor formation rate was 81.8%. The lymph node metastasis rate was 100%. The spleen metastasis rate was 63.6%. The renal metastasis rate was 45.5%. The abdominal implantation rate was 54,5%. The ascites formation rate was 54.5%;4.In group B, the nude mice were implanted with HGC27 tissue, the rate of gastric in situ formation was 100%. The rate of lymph node metastasis was 100%. The rate of spleen metastasis was 83.3%. the rate of renal metastasis was 66.7%. the rate of peritoneal implantation was 100%. the rate of ascites was 100%;5.5.The weight of nude mice in A and B groups showed a decreasing trend by time. the median survival time of A group was 38 days, and the median survival time was 30 days in group B;Conclusion:Nude mice model is more suitable than New Zealand white rabbits for the establishment of the animal model of lymph node metastasis in gastric carcinoma.In the animal model of nude mice, the models using cell suspension method has a longer median survival time and a lower rate of ascites formation, which is suitable for the study of lymph node metastasis mechanism of in gastric cancer. Models Using Tissue block has a high rate of abdominal cavity implantation and ascites formation, which is suitable for the study of the peritoneal metastasis mechanism in gastric cancer.
