Objective Using water extract of Cornus officinalis as the research object, based on the membrane separation technology, discusses the applicability of the Purification of anti-inflammatory components from Cornus officinalis, as the membrane separation technology in the field of modern Chinese medicine get more and more widely used to lay a certain foundation.Methods 1. Preparation and evaluation of water extract of Cornus officinalis:usingsmall ceramic membrane LNG-CM-101 laboratory equipment(aperture 0.8 μm、0.2 μm and 0.05 μm) and LNG-HFM-101 small hollow fiber membrane device(intercept molecular weight 50 kDa 、 10 kDa and 6 kDa) refined water extract in Cornus officinalis and optimize the preparation procedure through physical and chemical parameters, the content of Cornus officinalis total glycosides.2. We detect water extract of Cornus officinalis、microfiltrate with water extract of Cornus officinalis and ultrafiltrate with water extract of Cornus officinalis to find the influence of Cell proliferation to synovial Cell of fibroblasts(HFLS) with Cell Counting Kit- 8(CCK 8) method. Then testing different samples with interleukin(IL)- 1 beta and tumor necrosis factor(TNF)- alpha induced vascular endothelial growth factor(VEGF) and interleukin(IL)- 6 inhibitory effect, evaluation of membrane separation technology from the aspects of pharmacodynamics in vitro enrichment water extract of Cornus officinalis of saponins class anti-inflammatory component applicability.3.Water extract of Cornus officinalis、microfiltrate with water extract of Cornus officinalis and ultrafiltrate with water extract of Cornus officinalis and the total glucosides of paeony(TGP) to find the influence of the paw volume and weight by establishing the model of adjuvant arthritis rats. With ELISA method to detect adjuvant arthritis rats serum IL- 1, IL- 6, prostaglandin(PGE2) and the content of TNF alpha, through a microscope after H-E staining to observe the pathological slices,research and compare different samples of the therapeutic effect of adjuvant arthritis rats.Results 1.Inorganic ceramic membrane(0.05μm) showed better separation efficiency. The optimum operation pressure was 0.07 MPa 、 the temperature for filtration membrane was 40 ℃and the liquid temperature was 0.10 g·mL-1. The PS membrane with 10000 interception molecular weight had better separation efficiency.The optimum operation pressure was 0.1 MPa、the temperature for filtrationmembrane was 50 ℃and the liquid temperature was 0.13 g·mL-1. The water extract of Cornus officinalis after membrane refined, solution of turbidity, viscosity, particle size,color and other physical and chemical parameters have obvious improve, improve the clarity of the solution, reduce the suspended particles in the solution and polymer materials, and make effective components are well kept.Through comparing test characteristic fingerprint, the water extract of Cornus officinalis after refined membrane separation technology, its characteristics of fingerprint and concentrate has good similarity(> 0.998), indicated that membrane separation technology can effectively retain herbal medicine based on the overall effectiveness of the water extraction liquid of material group.2. In vitro pharmacodynamics experiment results show that the water extract of Cornus officinalis 、 microfiltrate with water extract of Cornus officinalis and ultrafiltrate with water extract of Cornus officinaliscan can effectively inhibit IL-1induced by excessive inflammatory cytokine VEGF, TNF-a, IL-6 and TNF-a induced excessive inflammatory cytokine VEGF, IL-6 Corni water extract by membrane refined enhances the its anti-inflammatory activity and the membrane ultrafiltration after anti-inflammatory activity was best.3. In vivo pharmacodynamics experiment results show that the water extract of Cornus officinalis 、 microfiltrate with water extract of Cornus officinalis and ultrafiltrate with water extract of Cornus officinaliscan can significantly down regulated in the rat model of adjuvant arthritis(AA) rats serum inflammatory cytokines IL-1, TNF-a, IL-6 and PGE2 improve inflammatory process of AA rats degree, which in ultra refined membrane solution experiments best results.Histopathology results show that the water extract of Cornus officinalis、microfiltrate with water extract of Cornus officinalis and ultrafiltrate with water extract of Cornus officinalis can inhibit synoviocytes of AA rats, proliferation of fibroblast cells and small blood vessels, to reduce the inflammatory cell infiltrationConclusion 1. Through the response surface method and membrane microfiltration membrane ultrafiltration process of Cornus officinalis water extracted liquid were optimized.2. By of different membrane separation technique to obtain the Cornus officinalis samples in vitro anti-inflammatory effects research, discussed the mechanism and inhibition of synovial cell excessive secretion of inflammatory cytokines related.3. through on different membrane separation technique to obtain the Cornus officinalis were anti-inflammatory effects in vivo study, found that Fructus Corni water extract refined membrane after anti-inflammatory effects enhanced by and with a molecular weight cut-off of 10 kDa hollow fiber membrane refining income effect of the ultrafiltrate best...
