Cell autophagy(autophagy) referred to as autophagy, is damaged organelles in the cell, such as aging of protein degradation, the digestion process, the vast majority of eukaryotic cells have this phenomenon.Generally speaking, the stability of cell autophagy helps to keep the stable state of the body, cell autophagy will be activated when stress, swallowed toxic or carcinogenic substance, inhibit the cells arise; But when excessive autophagy can damage cells and is the body disorder. Sports traing effect on regulating cell autophagy is a two-way street, if the intensity of exercise training is approptiate, then cell autophagy levels increase, some degradation due to the accumulation of metabolites in sports training and generate energy; If excessive movement, the excessive autophagy leads to cell organelles excessive degradation, protein, etc., such not only to maintain the stability of the function, on the contrary can damage cells, serious still can induce the death of cell autophagy. Research on cell autophagy more and more in recent years, fond that cell autophagy cells of the body stability, generation and metabolism, tissue reconstruction and synthesis is important, and to the body’s aging, organizing neural degenerative changes, gastrocnemius related diseases such as chronic disease has a lot of regulation function. At present, about the movement of gastrocnemius autophagy related research on the effects of gene expression is not a lot, and opinions vary. Smuder AJ and others through the experiment found that endurance exercise can enhance skeletal muscle antioxidant capacity; Whereas other studies found that endurance exercise and rat skeletal muscle autophagy level, there is no positive eddect. So, still need to further study of the correlation between motion and gastrocnemius muscle cell autophagy. In this paper, by studying the one-time exhaustive exercise on antioxidant levels of thr gastrocnemius muscle and cell autophagy and the effect of investigate the molecular mechanism of gastrocnemius muscle cell autophagy.Objective:acute exercise load model is established by exhaustion exercise once, detection of ICR mice gastrocnemius antioxidant function changes and autophagy related gene Atg5, Atg7, LC-3 and Beclinl expression level changes, one-time exhaustive exercise on autophagy, the influence mechanism of the gastrocnemius muscle, explore the one-time exhaustive on oxidation resistance of the gastrocnemius muscle oxygen levels and its cell autophagy and the effect of expected results for the reasonable arrangement of sprts training intensity and fatigue recovery to provide experimental basis.Methods:30 healthy male ICR mice,4-5 weeks, weight 25.17+/-2.46g, quiet were randomly divided into Control group and one-time exhaustive exercise group. Among them, the one-time exhaustive exercise group according to time divided into Oh,6h, 12h and 24h group. Mice fitness treadmill exercise,20 m/min speed, duration of 5min, trained for three days. Formal training to adapt at a speed of 20 m/min 30 min, then according to the following procedures:level 1 load:speed of 10 m/min, movement 15 min; Level 2 load:speed of 15 m/min, movement 15 min; Level 3 load:speed of 20 m/min, and then moving to exhaustion exercise group mice. Exhaustion standard: mice can’t maintain the preset speed, to stay in the end of the runway, electrical stimulation to drive. Standard instrument adopts Tecan Infinite M200 enzyme gastrocnemius T-SOD activity, Cu-ZnSOD activity, MnSOD activity, MDA content, T-AOC content. Real-time PCR determination of autophagy related genes of the gastrocnemius muscle in mice Atg5, Atg7, LC-3 and Beclinl mRNA expression level. Results:(1) compared with control group, immediately after exercise(Oh),6h Atg5mRNA expression level in mice has a rising trend,12h,24h has a downward trend, but there were no statistical significance(p> 0.05);(2) compared with control group, immediately after exercise(Oh),6h Atg7mRNA expression level in mice has a rising trend,12h,24h has a downward trend; In 12h has extremely significant difference(p< 0.01), the rest of the time points had no statistical significance(p> 0.05);(3) compared with control group, immediately after exercise(Oh),12h,24h LC-3mRNA expression level in mice has a downward trend,6h has a rising trend, but there were no statistical significance(p>0.05);(4) Compared with control group, immediately after exercise(Oh),6h,24h Beclinl mRNA expression level in mice has a rising trend,12h has a downward trend, In Oh,6h has extremely significant difference(p< 0.01), the rest of the time points has no statistical significance(p> 0.05);(5) Compared with control group, immediately after exercise(Oh),6h,12h T-SOD activity in mice and decline,24h and the control group; In 6h has significant difference(p< 0.05), the rest of the time points has no statistical significance(p> 0.05);(6) Compared with control group, immediately after exercise (0h),24h Cu-ZnSOD activity in mice has a rising trend,12h has a downward trend; In Oh,6h has significant difference(p< 0.05), the rest of the time points has no statistical significance(p>0.05);(7) Compared with control group, immediately after exercise (0h) MnSOD activity in mice has a rising trend, and has extremely significant difference(p<0.01); the rest of the time points has no change and has no statistical significance(p>0.05);(8) compared with control group, immediately after exercise(Oh),6h,12h,24h MDA content in mice has a downward trend, but there were no statistical significance(p>0.05);(9) compared with control group, immediately after exercise(Oh),6h,12h,24h T-AOC content are on the rise, and in 6h, 12h has significant difference(p<0.05), in 24h has extremely significant difference(p <0.01).Conclusion:(1) One-time exhaustion exercise activates the mice skeletal muscle autophagy related gene expression;(2)Skeletal muscle in mice after one-time exhaustion exercise is to enhance the antioxidant defense function.
