The paper on the "Peacock Princess" brand toothpaste with the Periplaneta americana extract chemical constituents. Chemical constituents of water-soluble fraction of ethanol extract of P.americana were isolated and purified by various chromatographic methods, including Silica gel, Sephadex LH-20 and RP-C18. Eight compounds were separated from P. americana and their structures were identified as 3,6-dimethylpiperazine-2,5-dione(1),tris(2-aminoethyl)amine(2),L-proline(3),D-valine(4),4-hydroxy-L-proline(5),L-leucine(6),L-alanine(7),Isoleucine(8) by spectral analysis including NMR and MS data. Among them, compounds 1~2 were obtained from the P.americana for the first time.This study shows that the "Peacock Princess" brand toothpaste with the P.americana extract valine(4), leucine(6), alanine(7) as the main ingredient. According to the experimental process, it disclosed that large-scale commercial production in amino acids and small molecules peptides separation by traditional silica gel does not have a good effect. Through the separate ways to explore to provide new ideas and methods for further study of the P.americana isolated constituents.Secondly, We compared the different extraction methods from P.americana with amino acid ingredient, experiments show that it’s difficult to effectively separate amino acids(or proteins) of a similar nature. we have try to test the proportion of preparation, compare different product content and components, the results show that a big difference. So the use of different process to purify or remove the element of specific categories, the effectiveness of different products can be obtained. It provides a new method for P.americana application.Finally, the medicinal sources of P.americana, new drug development, chemical constituents, pharmacological effects and content detection were summarized, such as research for the chemical composition of P.americana separation methods and provide reference to better explore the pharmacological activity.
