| According to the specific sequence of Salmonella enterica, Salmonella enteritidis and Salmonella typhimurium, amplification primers and sequencing primers were designed to establish the pyrosequencing method in this study. Target fragments were amplified by the specific primers of Salmonella enterica, Salmonella enteritidis and Salmonella typhimurium. Single-stranded templates were obtained from the PCR products and pyrosequencing were carried on with sequencing primers.The results showed that 24 strains of Salmonella enterica,16 strains of Salmonella enteritidis and 6 strains of Salmonella Typhimurium coming from different sources were complied 81bp,176bp and 131bp fragments,while the negative control strains were not amplified. Compared with BLAST, The following nucleotide sequence that could be completely matched with Salmonella enterica, Salmonella enteritidis, Salmonella typhimurium were GCCTGTGGGT ACTTCTCCTG CCAGTATAAT, CCTGTTGTCT GCTCACCATT CGCCAGCCAC, TACAACCGGA GTGCACATTA ATCCCGCAGC.The pyrosequencing method that showed a high efficiency and accuracy could be applied for the detection of Salmonella enterica, Salmonella enteritidis and Salmonella typhimurium in food. |
