Objective: By drawing materials of mouse embryos at different growth periods, investigate the transcription factor Brn3 a and Islet factor 1(Isl-1) co-expressed in the growth and development of the three periods in the trigeminal ganglion, Isl-1 transcription factor proliferation and Brn3 a transcription factor apoptosis, in order to study the growth of pulp nerves to provide new ideas and experimental theoretical projection. Methods: SPF level C57BL/6male and female mice(weighing 20±5g) were randomly assigned, breeders to 13.5days 15.5days 17.5days pregnant mice, mice were dissected embryonic head, each period has mutant and control, make frozen sections folliwing by immunofluorescence labeling Brn3 a 、 Isl-1, Brdu, Capcase3 neurons expressed, by semi-quantitative flourescence particles analysis and qualitative analysis of image. To compare the 13.5days, 15.5days, 17.5days in trigeminal ganglia Brn3 a and Isl-1 two kinds of transcription factors, Isl-1 proliferation expression and Brn3 a apoptosis expression. Results: In the trigeminal ganglia, Brn3 a and Isl-1 are co-expressed at 13.5days, 15.5days, 17.5days. Isl-1 expresses during the whole development stage, but few neurons are proliferating neurons, Brn3 a appears apoptosis of from 13.5days to 17.5days, but in the later stage Brn3 a are not so significant. Conclusions: 1) Brn3 a and Isl-1 positive neurons are decreasing during the development progress, indicating that Brn3 a and Isl-1 may play a role in regulating the upstream and downstream transcription factors during the growth and development, so as to control the pulp nerve growth projection. 2) Isl-1 is inactive at the three stages, suggesting it may play a role in the regulation of neural differentiation. 3) Brn3 a positive neurons are less in the three periods, which may develop into differentiated neuronal subtypes regulating differentiation and development of axons.
