Objectives:Aim to study the effect of estrogen levels and gender differences on blood pressure and the level of oxidative stress in different stage of renovascular hypertensive rats. Methods:Animal models were established respectively at different stages. According to the need of the experiment, parts of experimental animals were established to renovascular hypertensive rats model, the renovascular hypertensive rats model was established by 2K.IC method, and other parts of animals were ovarieetomized. Cardiac sonography, haemodynamics and the relevant indicators of oxidative stress level in myocardial and kidney tissue such as SOD、MDA、T-AOC、NO、anti superoxide anion determined respectively 16 and 32 weeks after operation. Results:The echocardiography results showed that 16 weeks after operation, compared with female 2KIC group LVIDd and LVPWd level were significantly reduced in female 2KIC ovarieetomized group; Compared with female 2KIC group and female 2KIC ovariectomized group, LVPWd level was increased significantly in male 2KIC group 32 weeks after operation,compared with female ovarieetomized group, LVSd level in female 2KIC ovariectomized group and LVIDd level in male 2KIC group were increased significantly. Compared with female 2KIC group, LVIDd level was decreased significantly in female 2KIC ovariectomized group. According to haemodynamic results,16 weeks after surgery, compared with female 2KIC group, SBP and DBP level both were reduced significantly in female 2KIC ovariectomized and male 2KIC group.32 weeks after surgery, compared with female ovariectomized group,SBP and DBP were increased significantly in female 2KIC ovarieetomized group, female 2KIC group and male 2KIC group. Compared with female 2KIC group, SBP level was reduced significantly in female 2KIC ovarieetomized group. Effect of estrogen on the antioxidant ability of kidney tissue in renovascular hypertensive rats:16 weeks after surgery, Compared with female 2KIC group, SOD、T-AOC level were increased in female 2KIC ovarieetomized group, SOD、T-AOC n anti O2- level was increased and MDA level was decreased in male 2KIC group.32 weeks after surgery,compared with female ovariectomized group, SOD and anti O2- level was decreased and MDA level was increased in female 2KIC group. Compared with female 2KIC group, SOD、T-AOC level were increased in female 2KIC ovariectomized group, SOD level was also increased and MDA decreased in male 2KIC group. Effect of estrogen on the antioxidant capacity of heart tissue in renovascular hypertensive rats:16 weeks after surgery,compared with female 2KIC group, SOD level was increased and eNOS、 MDA level were decreased in female 2KIC ovariectomized group, SOD、T-AOC、anti O2-level were increased and eNOS^ MDA level were decreased in male 2KIC group.32 weeks after surgery, compared with female ovariectomized group, SOD^ anti O2’level were decreased and MDA increased in female 2KIC group, compared with female 2KIC group anti O2-level was increased in male 2KIC group. Conclusion:Estrogen can promote the level of blood pressure and increase the level of oxidative stress in renovascular hypertensive rats, ovaries were removed and estrogen level below and improvement in severity, the mechanism maybe related to the influence on rennin angiotensin system.
