Objective: To study the situation of the key indexes ABP and StAr proteins expressed in the testicular tissue of abnormal Phlegmatic impotence rats, and the condition of testicular germ cells and leydig cells apoptosis,plus through the observing rat’s testicular structure to study the mechanism of this kind of rat model’ s accompanied oligospermia and the bio-synergy of the pharmacological effect. Method: A total of 90 normal mature male SD rats were enrolled in the study,and 10 of them were randomly chosen as the control group(N),other 80 were treated with spinach and coriander diet in a cold and humid environment until the model of abnormal Phlegmatic syndrome was established.Biological characteristics of rats were observed about 20 weeks until establishment of abnormal Phlegmatic abnormal Phlegmatic non-impotence group,then through APO erectile experiment and matching experiment,the rats were randomly divided into abnormal Phlegmatic syndrome with non-impotence rats(B1),medication intervention group of abnormal Phlegmatic syndrome with non-impotence group(B3), abnormal Phlegmatic syndrome impotence disease model group(A1), and medication intervention group of abnormal Phlegmatic syndrome with non-impotence group(A3). After2 weeks intervention:(1) Did HE staining to observe the group N and model groups morphological structure changes of testicular tissue and applied Electronic Speculum to observe its oon olutedseminiferoustubule;(2) Using immunohistochemical staining method to detect normal group and model group ABP and StAr proteins in the testicular tissue of rats under light microscopy;(3)Test the apoptosis level of leydig cell and all level germ celss and. Results:(1) HE staining testicular sperm cells morphological structure observation result showed,the testicular structure of group N was ordered; B1 and A1 group rats’ sperm cells were significantly less than the normal control group, the decrease in the number of leydig cells and sperm; B3 and A3 group germ cell wall atrophy and distorted into irregular shape, all level germ cells significantly less than the normal control group and decreased in the number of leydig cell(2) HE staining result showed, N group rat leydig cells assumes the circular or elliptic, full, cytoplasmic volume. B1 and A1group group rat leydig cells happened irregular shape, edema, darker, less. B3 andA3 group group of leydig cells have upward trend.(3) Electron microscopy(sem) results showed that N group of testicular tissue stroma cells, form a multilateral oval or round, abundant cytoplasm, cell membrane integrity.B1 and A1group all level germ cell was getting fewer and its structure was for some degree broken(4) Immunohistochemical(IHC)results show that the normal control group rats testicles StAr protein expression is significantly higher than other groups(P < 0.01). Abnormal Phlegmatic non-impotence group rats testicles St Ar protein expression significantly lower than the normal control group(P < 0.01).Abnormal Phlegmatic non-impotence medication intervention group group rats testicles St Ar protein expression is significantly higher than abnormal Phlegmatic non-impotence group group, model group and disease and disease drugs counterevidence group(P<0.01)but significantly lower than the normal control group(P<0.01). Abnormal Phlegmatic impotence disease medication intervention group testis St Ar protein expression group was obviously higher than that of disease model groups but significantly lower than the normal control group and abnormal Phlegmatic non-impotence medication intervention group group.(5) Immunohistochemical results showed that the normal control group rats testicles in ABP expression was significantly higher than other groups(P<0.01). ABP expression in abnormal Phlegmatic non-impotence group was obviously lower than that of normal control group. ABP expression abnormal Phlegmatic non-impotence medication intervention group group was obviously higher than that of abnormal Phlegmatic nonimpotence group(P<0.01) but lower than that of normal control group(P<0.01). Model group testicular disease and ABP expression significantly lower than the normal control group(P < 0.01) and the abnormal Phlegmatic non-impotence medication intervention group(P<0.01). Disease drugs anyway group rats testicles ABP expression significantly lower than the normal control group(P < 0.01) and the abnormal Phlegmatic nonimpotence medication intervention group(P < 0.01) but the disease is significantly higher than model group(P < 0.01). Abnormal Phlegmatic non-impotence group rats testicles in ABP expression and abnormal Phlegmatic impotence disease group rats testicles in ABP expression between amount of no statistical significance(P > 0.05).(6)TUNEL results showed that the normal control group in the rat leydig cells visibly shown scattered irregular single cell apoptosis. Abnormal Phlegmatic non-impotence group rat leydig cells apoptosis significantly more than normal group(P < 0.05).Abnormal Phlegmatic impotence medication group testicular happened the most leydig cell apoptosis. Abnormal Phlegmatic non-impotence medication intervention group leydig cells apoptosis has a decreasing trend(P < 0.05).(7) TUNEL method to detect apoptosis testicular sperm production, according to the fruit born B1 and A1 group sperm cells apoptosis significantly increased in the N group(P < 0.01), the yi wood sark apoptosis decline after intervention, B3 born sperm cells apoptosis number less than B1 group(P <0.01), more than N, but statistically significant difference(P > 0.05); A3 born sperm cells apoptosis number less than A1 group, more than N group, the difference was statistically significant(P < 0.01). Conclusion:(1) Uygur medicine phlegmatic impotence disease rat testis morphology structure change and living sperm cells apoptosis may be the pathological basis of essence of dysfunction.(2) abnormal phlegmatic type impotence disease rat testis reduced T, ABP, St Ar protein may be one of the important link of rat sperm production dysfunction.(3) The Yimusake tablet may influence by multiple drug targets for improving the structure of the testis morphology abnormal phlegmatic type impotence disease rat sperm production function changes.
