Root canal disinfection is the important step in the process of root canal treatment, also is the key to the success of root canal treatment. Laser disinfection technology is a hot spot in recent studies, dual wavelength laser disinfection is to use E r-YAG and N d:YAG to washing and disinfecting root canal, but at present the study of its sterilization effect is still less. ATP bioluminescence technique is a convenient and rapid detection method. It is applied in the environment, food, health care, etc. But in terms of root canal bacteria detection research, bacteria in rapid detection of clinical also lack volume method, and ATP has the advantages of this test in this paper to study the specificity and sensitivity of the ATP, and the use of ATP detection comparative dual wavelength laser disinfection method and the effect of the other methods.[Objective] Study the specificity and sensitivity of the ATP bioluminescence technique. Also this study is to compare the effect of double wavelength laser and sodium hypochlorite in disinfecting root canal.[Materials and Methods] The amount of E. faecalis was determined by serial dilution and plating on agar plates. Serially diluted microorganisms were subjected to the ATP assay. E. faecalis was treated with freshly prepared 1% NaOCl for 2.5,5, or 10 minutes; after which, NaOCl was neutralized with sodium thiosulfate. Bacterial cell viability was determined by culture as described previously. The ATP assay was performed on an aliquot of the treated sample. Collect 45 premolars from Dental hospital affiliated to Nanjing University. Removal of dental crowns, keep 10 mm in length.E. faecalis infected root canal model is set up. The tooth were divided into four groups of physiological saline group, dual wavelength laser group, neodymium laser group and sodium hypochlorite group. Before and after processing, we collected bacteria samples by3 sterile paper points. Recorded the amount of light of ATP bioluminescence technique.[Results] The luminescent signals, which were proportional to the amounts of ATP present, showed good correlations with the number of bacterial cells over a wide range for the bacterial species tested. Good correlations were obtained between the ATP readings and bacterial viability as determined by culture. There are Statistically differences between laser group and physiological saline group in the sterilization rate. There are Statistically differences between dual wavelength laser group and sodium hypochlorite group in the sterilization rate. There are no statistically differences between dual wavelength laser group and neodymium laser group in the sterilization rate.[Conclusion] The ATP detection method can be used as a rapid tool to determine the presence of E. faecalis. The effect of Double wavelength disinfection is better than that of sodium hypochlorite flushing.
