Objective:Our aim is to evaluate the expression levels of TRPP2, COX-2 and MMP-14 protein in laryngeal carcinoma tissues, to analyze the relationship between the overexpression of TRPP2, COX-2 and MMP-14 protein in laryngeal carcinoma and their clinical parameters.Furthermore, we figure out if TRPP2, COX-2 and MMP-14 protein expression intensity were related to the prognosis of patients with laryngeal cancer.Methods:We collected 100 cases of surgical treatment of cancer patients with laryngeal carcinoma and 42 cases adjacent tissue specimens from January 2008 to December 2010 in the Department of Otolaryngology Head and neck surgery of the First Affiliated Hospital of Medical University Of Anhui.The expression was detected by immunohistochemical method of TRPP2, COX-2 and MMP-14 protein in laryngeal carcinoma tissues and adjacent tissue.We calculate the OD value by using Image-Proplus 6 software and use Epi Data3.0 and SPSS18.0 software to establish a database and analyzed statistically.Result:In this study,our results showed that the expression intensity of TRPP2, COX-2and MMP-14 protein in laryngeal carcinoma tissues were higher than the expression in adjacent tissue.There was no significant difference In different age groups(60 years as the boundary), smoking group, alcohol group(200ml/d the boundary),between pathological differentiation group and glottic and non glottic group.In clinical stage I +II of laryngeal carcinoma, the expression of TRPP2,COX-2 and MMP-14 protein were weaker than the expression in stage III and IV of TRPP2,COX-2 and MMP-14 protein in laryngeal carcinoma(P<0.05).Moreover, increased TRPP2,COX-2 and MMP-14 expression was strongly associated with lymph node metastasis.TRPP2, COX-2 and MMP-14 protein expression intensity was higher than the 50,70,85 percentile level of the patients, whose survival time was shorter than that of TRPP2, COX-2 and MMP-14 protein expression intensity was lower than the 50,70,85 percentile level.Pearson correlation analysis showed COX-2, MMP-14 has a significant positive correlation between each other(r=0.244,P=0.014),COX-2, TRPP2 has a significant positive correlation between each other(r=0.227,P=0.023).But the correlation between MMP-14 and TRPP2 is not obvious(r=0.190,P=0.059>0.05).Conclusion:1.TRPP2, COX-2 and MMP-14 expression intensity was significantly higher than that of adjacent tissues in laryngeal carcinoma, suggesting that TRPP2, COX-2and MMP-14 protein overexpression was related to the occurrence of laryngeal squamous cell carcinoma.2.TRPP2, COX-2 and MMP-14 protein in the late stage of T, clinical stage for IV+III and have lymph node metastasis were strong expression.This indicates that the expression of TRPP2, COX-2 and MMP-14 protein was related to the invasion and metastasis of laryngeal carcinoma.3.Overexpression of TRPP2,COX-2 and MMP-14 protein in laryngeal cancer patients survival rate were significantly lower than that of TRPP2,COX-2 and MMP-14 low expression intensity of patients,and suggests that the expression of TRPP2, COX-2 and MMP-14 protein were related to the prognosis of the patients.4.COX-2, MMP-14 has a significant positive correlation between each other.COX-2,TRPP2 has a significant positive correlation between each other.But the correlation between MMP-14 and TRPP2 is not obvious.The specific reasons need to be further studied.
