Changes Of Intestinal Epithelial Barrier In Hepatic Fibrosis Rats And Protective Effects Of Hypericum Perforatum Extract

Objective To investigate the protective effect of Hypericum perforatum extract(HPE) on intestinal barrier in hepatic fibrosis ratsMethods Fifty SD rats were randomly divided into five groups: normal group; model group; HPE(25,50,100 mg/kg) groups. Different doses of HPE(25, 50, 100 mg/kg) and model group were given to subcutaneous inject with 50%CCL4/peanut oil and normal group was given to subcutaneous inject with the same peanut oil,twice a week. Different doses of HPE(25, 50, 100 mg/kg) were given to rats in HPE groups by gavage. After 12 weeks later, rats were weighed and sacrificed. The liver and spleen index were calculated, the level of IV-C, the level of ZO-1, occludin, GFAP, S100 B and GDNF in intestinal tissues were tested. Additionally, the hepatic pathological changes were observed.Results Successful liver fibrosis model was established. Compared with nomal group, hepatocellular of model group blockbuster necrousis,and structure were disordered; intestinal mucous of jejunum ileum and colon were fused;the vilus were faulted; intestinal epithelial cells were necrousised and the permeability of intestinal epithelial increased. Compared with nomal group, the level of IV-C,ZO-1, occludin and GDNF were signifieantly lower than normal group(P<0.05); the level of GFAP, and S100B werer signifieantly higher than normal group(P<0.05). Compared with model group, the content of IV-C in blood serum was decreased; the content of GFAP, S100 B in intestinal tissues were decreased, the content of ZO-1, occludin, GDNF in intestinal tissues were increased in HPE groups. The pathologielesions were found in the intestinalmueosa and the liver tissue of the model group and the iniestinal villus height shorten sign ifieantly, compared with the normal group.Conclusion Liver fibrosis associated with the injury of intestinal barrier function and there were no; significantly differences among jejunum ileum and colon. HPE has protective effect on intestinal barrier.The mechanism may be related to the activation glial cells in the regulation of the intestinal epithelial barrier,;the mechanism of HPE mybe related to activate glial cells and control intestinal barrier.
