Background & aim:In a variety of Cathepsin S(CTSS) study,we found that CTSS in a number of malignant tumors increased in expression such as liver cancer, stomach cancer, bladder cancer, etc. CTSS involved in tumor angiogenesis, increased tumor invasiveness, metastasis,is one of the factors of tumor progression and metastasis;We also found that melittin have anti-tumor properties, inhibit the growth of tumors and can induce tumor cell apoptosis.We have established the silence CTSS MHCC97 H stable cell lines,and use melittin intervention it to inquiry silence CTSS and the effects of melittin on cell lines which of silenced CTSS’s situation.We through the establishment of silence CTSS MHCC97-H tumor model in nude mice, using melittin intervention to discussion whether silence CTSS and melittin have the sameeffect in vivo.Methods:Prepared by the silence CTSS MHCC97-H cell line(marked silence CTSS cell lines is PLVT1150, table CTSS normal cell lines referred to as PLVT8),and Using flow cytometry detect cell lines interventioned by different concentrations of melittin,and the effect of melittin on Apoptosis about MHCC97-H cells.To establish silence CTSS MHCC97-H tumor-bearing nude mice model.Use melittin intervention it and investigate the status of tumor growth in nude mice to discussion mechanism of silence CTSS and melittin in vivo.Using immunohistochemical expression caspase3, clevedcaspase3 in tumor-bearing nude mice model,to explore melittin and silence CTSS cause tumor cell apoptosis’ s pathway.Results:We detected PLVT8 and PLVT1150 cells’ s apoptosis after melittin(0、4、8ug/ml)intervention 30 minutes by flow cytometry. The experiment was repeated 3 times. After melittin(0,4,8 ug / ml)intervention the two cells group apoptosis rate were(18.9%±2.1vs.4.3%±1.9、36.7%±2.2vs.15.3%±1.7、53.0%±1.5vs.28.9%±2.2). In nude mice model after the silencing CTSS,Melittin collaborative silencing CTSS can significantly inhibit tumor growth in animal models.Its showed that melittin may have a role in promoting apoptosis of MHCC97-H cell. In immunohistochemistry experiments, caspase3 expression decreased, while cleved-caspase3 expression increased.Conclusion:Lentiviral-mediated silencing CTSS promote MHCC97-H apoptosis,and melittin can promote apoptosis of MHCC97-H cell after silence CTSS;Combined silence CTSS and melittin treatment may add a new research direction for biological treatment of hepatocellular carcinoma.
