Objective To explore the expression of CCR6-CCL20 axis and CCR4-CCL22 axis in circulating of SLE patients, investigate the relation between these two axes and clinical characteristics of SLE.Method 22 SLE patients and 20 Healthy controls were chosen to attend the research according to the inclusion and exclusion criteria. Record the clinical data of patients at pretreatment and post-treatment. The level of CCL20 and CCL22 were assayed by ELISA, and the expression of CCR4 and CCR6 were assayed by FACS analysis. Comparative and correlation analysis was conducted by SPSS 19.0.Result (1) CCR6-CCL20 axis:The CCL20 levels in SLE patients were significantly higher than those in healthy controls(P<0.05). Higher level of CCL20 was found in primary SLE patients with kidney lesion and serositis and decrease after treatment (P<0.05). The CCL20 levels in SLE patients had correlation with SLEDAI and C3, ds-DNA (all P<0.05). The expressions of CCR6 had no significantly difference between SLE patients and healthy controls and had no obvious change after treatment. The expressions of CCR6 had no correlation with SLEDAI and C3, ds-DNA (all P>0.05). (2) CCR4-CCL22 axis:The CCL22 levels in relapsing SLE patients were significantly lower than those in healthy controls(P<0.05). Lower levels of CCL22 were found in primary SLE patients with kidney lesion and serositis (P>0.05). The levels of CCL22 in SLE patients decreased after treatment. But CCL22 level had no correlation with SLEDAI and C3, ds-DNA (all P>0.05). The expressions of CCR4 in SLE patients were significantly higher than those in healthy controls(P<0.05). The levels of CCR4 in SLE patients with kidney lesion and serositis were higher than those in SLE patients with serositis only or without kidney lesion and serositis (P<0.05). The levels of CCR4 in relapsing SLE patients decreased after treatment(P<0.05). And the expressions of CCR4 had correlation with SLEDAI and C3, ds-DNA (all P<0.05).Conclusion (1) CCR6-CCL20 axis and CCR4-CCL22 axis had correlation with the occurrence, development and prognosis of SLE. (2) The expressions of CCR4 and levels of CCL20 might become the index for the activity of SLE.
