In vitro fertilization technology is significance that some endangered animals, and varieties of livestock breed offspring continue numerous, international exchange and the variety resources saving, its biggest advantage is to break the time and space limit. But in vitro fertilization rate is low, so the purpose of this experiment is to through optimizing conditions improving the development of the vitro fertilization rate. Optimizing the egg time, capacitation and sperm egg incubation time and using DCFDA dyeing tests the level of ROS and immunofluorescence detection FOXO3a expression to examine the effect of active oxygen for embryonic principle. Research results show that the mining eggs at 13 h, capacitation time 1.5 h, sperm egg incubation time 6 h, in vitro fertilization (ivf) in mice results is best; After H2O2 treatment in different stages of the embryo FOXO3a expression were significantly higher than the control group. Above all through the time to collect eggs and capacitation time and sperm egg incubation time optimization to improve the vitro fertilization rate, and through detection of FOXO3a protein content further illustrate FOXO3a involved in oxidative stress reaction.
