Objective: To explore the effects and mechanisms of microRNA-101(miR-101) suppresses bladder cancer by targeting EZH2. Methods: 1. miR-101 was quantified in human bladder cancer tissue and adjacent nontumor tissues by quantitative Real-Time PCR; 2. We used immunohistochemical(IHC) method to investigate EZH2 expression in bladder cancer and adjacent nontumor tissues through tissue chip; 3. RT-PCR, Western blot were used to detect the expression of EZH2 in bladder tumor and corresponding tissues; 4. The software for predicting mi RNAs: mi Rnada, PicTar and TargetScan were used to predict the potential target genes of miR-101; 5. To prove miR-101 has the combine target spot with EZH2 3’- UTR by dual-luciferase reporter gene system. Results: 1. miRNA-101 was found had a lower lever in bladder cancer compared with its adjacent nontumor tissues by quantitative Real-Time PCR, indicating that miR-101 may inhibits the development of bladder cancer; 2. The IHC, RT-PCR, Western blot analysis verified that EZH2 expression was significantly increased in more than 70% of bladder tumor compared with their nontumoe parts, proving that EZH2 may promoting the progress of bladder cancer; 3. Through the predicting software and dual-luciferase reporter gene system we found miR-101 has the regions combine with EZH2 3’- UTR which makes miR-101 regulate EZH2 gene directly.Conclusions: EZH2 was negatively regulated by miR-101 to restrain the progress of bladder cancer.
