Objective: By exploring the distribution characteristics of the quinone oxidoreductase 1(quinone oxido-reductase 1, NQO1) gene C609 Tpolymorphism in Guilin area of female population. To investigate the relationship between NQO1 C609 T polymorphisms and genetic susceptibilities, combination with the clinicopathological characteristics of breast cancer patients, to analysis the relationship between NQO1 C609 T polymorphisms and clinicopathological characteristics of breast cancer,in order to provide a scientific basis for clinical practice and individualized treatment. Methods: By using case-control study of hospital-based collection, we collected 295 cases of female breast cancer patients who have confirmed by pathology in affiliated Hospital of Guilin Medical College from Oct. 2013 to Mar. 2015 as the experimental group. Meanwhile, we also collected 232 cases of healthy female in department of medical center, affiliated Hospital of Guilin Medical College from Jan. 2014 to Dec. 2014 as the control group, then collected all the research object of peripheral blood samples. By using TaqMan MGB fluorescence quantitative polymerase chain to analysis the genotypes of NQO1 gene C609 T. At the same time, detecting the part,pathological type, hostologic classification, size, TNM staging, ER, PR, HER-2, P53 and Ki-67 of the breast cancer patients, detecting the sampling balance of two groups by Hardy-Weinberg equilibrium test. All experimental date were analyzed in SPSS 18.0 statistical software. Apply chi-squared test and multivariate Logistic regression to analysis the genotypes distributed difference in the two groups, and the relationship of the three genotypes and clinicopathologic parameters was also analysed.Results:(1) There were no statistically differences in the age, nation and menopausal status in the 295 breast cancer patients and 232 control groups.(2) The Hardy-Weinberg equilibrium testing result of the NQO1 three genotypes(CC,CT,TT) in the patient group was(χ2=0.046,P=0.831), the Hardy-Weinberg equilibrium testing result of the NQO1 three genotypes(CC,CT,TT) in the control group was(χ2=0.064, P=0.801).They all haven’t the statistically significant.(3) In the control group, the frequencies of CC, CT, TT genotypes were 37.1%, 48.3% and 14.6%, in the case group, the frequencies of CC, CT, TT genotypes were 27.1%, 50.5% and 22.4%, the difference between two groups were statistically significant(χ2=8.289,P=0.016).(4) The frequencies of TT genotype in case group was higher than the control group. So the risk of breast cancer was higher in TT genotype carriers compared with CC genotype carriers(OR=2.087,95%CI:1.248~3.488).(5) The frequencies of the C allele and T allele showed a significant difference between patients with breast cancer and health controls(P=0.004), and the risk of breast cancer was higher in T allele carriers compared with C allele carriers(OR=1.435,95%CI:1.212~1.837).(6) The CC, CT genotype carrier compared with TT genotype carrier in the breast cancer patient, the lymph nodes metastasis was 56.3%, 50.0%, the difference between the two groups have statistical significance(P=0.046). The positive rate of Estrogen Receptor was 70.2%, 55.0%, the difference between the two groups have statistical significance(P=0.014), The positive rate of Progesterone Receptor was 61.9%, 47.5%, the difference between the two groups have statistical significance(P=0.026). Further analysis by multivariate Logistic regression, we conclude that the risk of breast cancer lymph nodes metastasis was higher in CT, TT genotype carriers compared with CC genotype carriers(OR=2.814,95%CI:1.404~5.693).Conclusion: NQO1 C609 T polymorphisms may increase breast cancer risk and correlated to ER, PR, lymph nodes metastasis, which suggest NQO1 C609 T polymorphisms may be a potential biomarker for poor prognostic evaluation of breast cancers.
