Objective: The purpose of this study was to observe the effect of receptor tyrosine kinases(AXL) on gallbladder cancer cells proliferation, by the same time to explore the mechanism during the procedure.Methods: 1.Immunohistochemical method was used to detect the expression of AXL in the gallbladder carcinoma tissue, then weanalyzed the significance of AXL positive expression with clinical data.2.After interfered the AXL by using RNA interference(RNAi), we detected the interference efficiency by using Western Bot,by the same time,we Selected the highest interference efficiency of a follow-up experiment interference fragment.3.Cell migration and invasion capability in GBC-SD was detected by Wound scratch assay, MTT and Transwell.4. The expression of PI3K/AKT pathway related proteins was detected by using Western Blot.Results: 1.By using Immuno histochemistry we found the expression of AXL in gallbladder carcinoma was significantly higher than that in chronic cholecystitis, and the positive expression of AXL was closely related to lymph node metastasis and tumor stage in patients with gallbladder carcinoma2.After interferening the AXL by using RNA interference(RNAi), the migration and invasion capability of gallbladder cancer cell was significantly lower compared with the cells without interference(P<0.05),the expressions of PI3 K and p-mtor were significantly lower than before(p<0.05).Conclusion: Keceptor tyrosine kinase- AXL could effect the proliferation of gallbladder cancer cells,which was significantly decreased after interferening. its role was dependent on the activation of protein associated with PI3 K.Development of their inhibitors were expected to improving the poor prognosis of patients with gallbladder cancer.
