ObjectiveTo observe the therapeutic effect and mechanism of Semen Litchi effective constituentson on nonalcoholic fatty liver disease in rats.MethodsNAFLD model of rats was induced by high fat diet for 8 weeks. Then model rats were randomly divided into 4 groups:model group, SLEC high-dose group(1.48g·kg-1·d-1), SLEC low-dose group(0.74g·kg-1·d-1), metformin grop (100mg·kg-1·d-1), and blank group was established as control group. All rats were fed with normal feedstuff. After medication for 4 weeks, triglyceride(TG), cholesterol (CHOL), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C), alanine aminotransferase (ALT), aspartase aminotransferase (AST)and fasting blood-glucose (FBG) content in the serum was detected by automatic biochemical analyzer. Non-esterified fatty acid(NEFA) content in the serum was detected by colorimetry. Serum insulin content was detected by radioimmunoassay (RIA). Homeostasis model of assessment for insulin resistence index (HOMA-IR) and insulin sensitivity index(ISI) were calculated to estimate the levers of insulin resistance(IR). TG, CHOL, superoxide dismutase(SOD), malondialdehyde (MDA) content in liver tissue were detected respectively by methods of glycerol phosphate oxidase-peroxidase (GPO-PAP), cholesterol oxidase-peroxidase (COD-PAP), water soluble tetrazolium(WST-1),thiobarbituric acid(TBA). Pathological change of liver tissue was observed by HE staining and Oil Red 0 staining. The sterol-regulatory element binding protein-lc (SREBP-lc) mRNA expression levels in liver tissue were detected by reverse transcription polymerase chain reaction (RT-PCR). The SREBP-lc protein expression levels in liver tissue were detected by western blot.ResultsCompared with the model group of rats, the content of TG, LDL-C, NEFA and liver lipid was significantly decreased (P<0.05); SOD content in liver tissue increased significantly (P<0.05); HOMA-IR decreased and ISI increased(P< 0.05), in SLEC high-dose group. The degree of hepatic steatosis was alleviated obviously in SLEC high-dose group. Compared with the model group, The mRNA and protein expression levers of SREBP-1c in SLEC high-dose group and SLEC low-dose group were obviously down-regulated (P<0.05).ConelusionSLEC showed certain therapeutic effect on hepatic steatosis in NAFLD rats, which might be related to regulating lipid metabolism, decreasing the content of NEFA, improving IR and oxidative stress and down-regulating the expression of SREBP-lc mRNA and protein.
