Effects Of Electroacupuncture On 6-OHDA-induced Parkinson’ Gastrointestinal Function

Objective:This study was to observe the changes of the diet, body weight, the amount of feces of 12h, fecal moisture percentage, the stomach retention rate, the tyrosine hydroxylase (TH) in the substantia nigra and the neuronal nitric oxide synthase (nNOS) in the substantia nigra, gastric antrum and proximal colon areas of the 6-OHDA induced Parkinson’s disease rats, define the related mechanisms of the gastrointestinal disorder symptoms of the Parkinson’s disease. What’s more we also observed the therapeutic effect of early intervention of electroacupuncture, late intervention of electroacupuncture and drug on gastrointestinal disorder symptoms of Parkinson’s disease as well as the change of TH and nNOS, explored the impact of electroacupuncture treatment on gastrointestinal disorder symptoms of Parkinson’s disease and pathological process of Parkinson’s disease.Methods:1.We injected the 6-OHDA or the saline containing 0.02%vitamin C into the substantia nigra area, using the brain stereotaxic instrument, to establish the model of Parkinson’s disease or the sham operation group.2. We observed the diet, body weight, the amount of feces of 12h, fecal moisture percentage, the stomach retention rate and the level of tyrosine hydroxylase (TH) in the substantia nigra and the neuronal nitric oxide synthase (nNOS) in the substantia nigra, gastric antrum and proximal colon areas of the normal group, the sham operation group and the model group.3.After the treatment of electroacupuncture or drug, we observed the diet, body weight, the amount of feces of 12h, fecal moisture percentage, the stomach retention rate, the level of tyrosine hydroxylase (TH) in the substantia nigra and the neuronal nitric oxide synthase (nNOS) in the substantia nigra, gastric antrum and proximal colon areas of the normal group, the sham operation group and the model group.Results:1.In terms of diet and body weight, the normal group and sham operation group had no statistical significance(P>0.05);the diet and body weight of 6-OHDA induced Parkinson’s disease group were lower than normal group or sham operation group, and had statistical significance (P<0.05).2.1n terms of the amount of feces of 12h, normal group and sham operation group had no statistical significance (P> 0.05); the amount of feces of 12h was decreased 31.1%compared with normal group in 6-OHDA induced Parkinson’s disease group,decreased 33.4%compared with sham operation group,and all had statistical significance (P< 0.05).3.In terms of thesolid residue rate in the stomach, there was no markedly differences between normal group and sham operation group(P>0.05); thesolid residue rate in the stomach of 6-OHDA induced Parkinson’s disease group was much higher than that of normal group and sham operation group (P< 0.05),increased 35.7%compared with normal group and increased 38.7%compared with sham operation group.4.Compared the therapeutic effects among the early electroacupuncture group, the late electroacupuncture group and the drug therapy group, we found that in terms of the diet and weight growth rate, the early electroacupuncture group is significantly higher than model group, and the differences were statistically significant (P< 0.05).What’s more, there were no significant differences between late electroacupuncture group and model group(P> 0.05), which also had no markedly differences with drug group(P> 0.05);The amount of feces of 12h, fecal moisture percentage and the stomach retention rate of early electroacupuncture group had no statistically significant differences while compared with model group(P< 0.05); the amount of feces of 12h in late electroacupuncture group was markedly different from that of normal group(P< 0.05), the fecal moisture percentage and the stomach retention rate in late electroacupuncture group had the trend to become normal; the amount of feces of 12h in drug group was much higher than model group(P< 0.05), the stomach retention rate of drug group was significantly higher than normal group (P< 0.05).5, in terms of the substantia nigra tyrosine hydroxylase (TH)’mean gray value(MGV), the comparison between normal group and control group had no statistical significance (P> 0.05); the model of Parkinson’s disease induced by 6-OHDA compared with normal group and sham operation group was increased, and had statistical significance (P< 0.05). After treatment, the substantia nigra tyrosine hydroxylase (TH) of the early electroacupuncture group obviously returned to normal, compared with model group was statistically significant (P< 0.05); the late electroacupuncture group compared with model group had significant difference (P< 0.05), drug group compared with model group had no statistical significance (P> 0.05), visibly the late electroacupuncture group had a better treatmental effect than the drug group.Compared TH cell number of per unit area in the substantia nigra positive area:control group and normal group is no statistical significance (P> 0.05), model group compared with normal group, control group had significant difference (P< 0.05); Early electric acupuncture group compared with model group had significant difference (P< 0.05), late electric acupuncture group compared with model group has no statistical significance (P> 0.05), drug group had the same result with late electric acupuncture group.6, In terms of the level of substantia nigra neurons nitric oxide synthase (nNOS), comparison between normal group and control group had no statistical significance (P> 0.05); 6-OHDA induced Parkinson’s disease model group was lower than normal group and sham operation group(P< 0.05). In the region of the gastric antrum and proximal colonic type,the level of neuronal nitric oxide synthase (nNOS) had no differences between normal group and sham operation group(P> 0.05); the level of neuronal nitric oxide synthase (nNOS) in the region of the gastric antrum and proximal colonic type of 6-OHDA induced Parkinson’s disease model group was higher than normal group and sham operation group(P< 0.05).After treatment, neuronal nitric oxide synthase (nNOS) in the areas of the substantia nigra, gastric antrum and proximal colonic type of the early electroacupuncture group had statistically significant differences while compared with model group(P> 0.05), which was obviously returning to normal; nNOS in the substantia nigra and proximal colon area of later electroacupuncture group had statistically significant difference when compared with model group (P< 0.05); compared with model group, drug group had no significant differences (P> 0.05).7> In terms of horizontal motion path, comparison between normal group and control group had no statistical significance (P> 0.05); 6-OHDA induced Parkinson’s disease model group was lower than normal group and sham operation group(P< 0.05). After treatment, early electroacupuncture group had statistically significant differences while compared with model group(P< 0.05), which was obviously returning to normal; later electroacupuncture group had statistically significant difference when compared with model group (P< 0.05); compared with model group, drug group had no significant differences (P> 0.05), later electroacupuncture group had statistically significant difference when compared with early electroacupuncture group (P< 0.05),the same to drug group.Conclutions:1.6-OHDA induced Parkinson’s disease model rats has gastrointestinal disorder symptoms, including the significantly decrease of diet, weight and the amount of feces of 12h, significantly slow of gastric emptying, rise of gastric solid residue rate.2.6-OHDA induced Parkinson’s disease model rats have gastrointestinal disorder symptoms, which may be associated with changes in nitric oxide level.3. The electroacupuncture has significant regulating effects on 6-OHDA induced Parkinson’s disease model rats.The regulating effects of electroacupuncture is better than drug, and the electroacupuncture treatment could improve the degradation of dopamine function of Parkinson’s disease; Early intervention of electroacupuncture has significantly better regulating effects than later intervention of electroacupuncture, maybe the effect was linked with nNOS.
