ObjectiveTo disscuss the material basis and the scientific connotation of cold-dampness syndrome ofrheumatoid arthritis.MethodRandomly to divid SD rats into the normal group,the model group,positive medicine group and NBTL group.Rats were used to built collage-induced arthritis animal models. Different groups were given corresponding drugs or distilled water for 28 days after successful model built. Body weight, Arthritis Index (AI value), and paw swelling were monitored to evaluate the CIA Model and efficacy of medicine.During the experiment, the blood samples were taken from the orbit and frozen for several times in order to study the experimental of metabolism in the later stage. We chose to observe jointspathological changes by ankle joint.Result1. Before medication-interventing,(before the intervention, the experiment for 0 days), We found that AI mean value in various model group is greater than 2, the AI value baseline in rats from various module group showed no significant difference (P>0.05) by macroscopic observation.2. During the intervention process, the New Bitongling could reduce the AI value and paw swelling caused by inflammation.However, there was only the difference between the time-course effect. There was no significant difference in the overall efficacy.Pathological HE staining shows that the New Bitongling can alleviate the synovial cells of rat ankle joint swelling and proliferation, inflammatory cell infiltration, pannus formation.3.By using the LC-MS, we can clearly distinguish the metabolism difference between before and after successful model, as well as the metabolism changes during the intervention. Further construction of the OPLS-da model, the model parameters were R2Y=0.98, Q2= 0.92, and we selected 10 abnormal biological metabolites, included creatinine, nicotinamide, phenylacetylglycine, palmitic acid, tryptophan, leucine and a variety of lipids. Creatinine, nicotinamide, benzene acetyl glycine, leucine, tryptophan acid, and lipids showed a downward trend in the plasma of the rats in the model group.Palmitic acid, LPC (18:0), LPC (20:4) were up-regulated in model group.For most of the abnormal metabolites, the regulatory effect of the Tripterygium is stronger than that of the New Bitongling, but for the leucine and tryptophan, the New Bitongling showed obvious advantages.Conclusion1. The New Bitongling could reduce the AI value in CIA rat, minize ankle swelling of CIA rats, inhibit articular pathological damage, which indicates that the new Bitongling different dose group can reduce joint inflammation, bone loss, alleviate RA bone destruction in a certain extent.2. LC-MS can clearly distinguish the metabolic pattern of normal rats and model rats, themetabolites changes during the intervention can be presented.3. The metabolic disorder of CIA rats was presented on the change of creatinine, nicotinamide, phenylacetylglycine, palmitic acid, tryptophan, leucine and a variety of lipids.4. The New Bitongling has a better regulation effect on the metabolism of the disorder in the plasma of CIA rats, especially the regulation of tryptophan and leucine.5. Rheumatoid arthritis of cold-dampness syndrome pathogenesis may be related to the IDO/TTS lubricious ammonia acid metabolic pathway and leucine disorders.
