Objective:The action mechanism of Qingfei Oral Liquid in treating infection of respiratory syncytial virus(RSV) is not clear deeply. From the aspect of sustaining Treg/Th17 type cytokines balance, this study aims at detecting the action mechanism of Qingfei Oral Liquid by examining the expression of Interleukin(IL)-10 and Interleukin (IL)-17 in mice’s lung tissue infected by RSV, and expression level of Treg and Th17 in mice’s spleen tissue before and after treatment.Method:Fifty BABL/c mice were randomly divided into 5 groups:control group, model group, Ribavirin group, Qingfei Oral Liquid low dose group(1.33g/ml) and high dose group(4g/ml).Control group was given intranasal drop of equal quantity and isosmotic saline solution, meanwhile other groups were modeled with RSV(long strain) virus. After activating virus for 48h, all groups accepted gavage once per day. Control group and model group were given 0.9% isosmotic saline solution, and high dose group, low dose group and Ribavirin group were given 4g/ml Qingfei Oral liquid,1.33g/ml Qingfei Oral liquid and 0.0025g/ml Ribavirin respectively. Mice were put to death after 72h, and 5 mice were selected from each group to take pathologic analysis for lung tissue, while the rest were used to examine the expression levels of IL-10 and IL-17 in bronchoalveolar lavage fluid by the method of ELISA. Four mice were selected to examine the expression levels of Treg and Th17 in spleen tissue by flow cytometry(FCM).Result:Compared with model group, pathologic change scores of high dose group, low dose group, Ribavirin group and control group were decreased(P<0.01), meanwhile IL-10 expressions in these four groups (4were obviously increased(P<0.01) by comparing with model group, and IL-17 expressions, compared with model group, decreased apparently(P<0.01).Expressions of IL-10 and IL-17 in high dose group and low dose group were decreased and increased respectively by comparing with Ribavirin group (P<0.05). Expression levels of Treg and Th17 in high dose group, low dose group and Ribavirin group rised up by comparing with model group(P<0.01). Compared with Ribavirin group, expression of Treg and Th17 in both Qingfei Oral Liquid groups showed the statistic difference.(P<0.05)Conclusion:Qingfei Oral Liquid can regulate the balance of Treg/Th17 and increase the expression of IL-10 in lung tissue of infected mice, which further proves that there are effects of Qingfei Oral Liquid on treating RSV viral pneumonia.
