Objective:This experiment has observed the effect of Jian Pi Hua Yu Decoction on the proliferation in human gastric cancer cell line SGC7901/DDP and possible mechanisms of drug-resistance reversal. We can further explore the role of traditional Chinese medicine in terms of anti-tumor, then to provide new ideas for comprehensive treatment of tumors.Methods:1. Drug-resistance of SGC7901/DDP was evaluated by MTT method.2. The cytotoxicity of Jian Pi Hua Yu Decoction was evaluated by MTT method.3. Flow cytometry was used to measure the promotion of apoptosis.4. Bcl-2, Bax, PTEN mRNA expression in SGC7901/DDP cells treated with or without Jian Pi Hua Yu Decoction was detected by RT-PCR.5. The expression of apoptosis associated proteins Akt, P-Akt, PI3K, NF-κB p65 were detected by Western blot.Results:1. SGC7901/DDP has significantly lower sensitivity to cisplatin than SGC7901, indicating drug-resistance.2. With the increase of drug concentration and time, the inhibitory effect on the growth of SGC7901/DDP cells also increased.3. Apoptosis rates and sensitivity to cisplatin of SGC7901/DDP cells induced by Jian Pi Hua Yu Decoction in a dose-dependent manner.4. Jian Pi Hua Yu Decoction decreased the mRNA expressions of Bcl-2 mRNA and increase Bax、PTEN of SGC7901/DDP cells.5. The expression of Akt, PI3K, NF-κB p65 were down-regulated by Jian Pi Hua Yu Decoction.Conclusions:Jian Pi Hua Yu Decoction inhibit the proliferation of human gastric cancer cell line SGC7901/DDP, induce this cell apoptosis and reverse the drug resistance. The possible mechanisms may be related to the regulation of PTEN/PI3K/AKT/NF-κB pathway.
