| ObjectiveLung cancer is the top list of cancer-related cause of death,among them 85% are NSCLC. Curcumin is the main extract of turmeric rhizome,It harbours wide range of pharmacological activity,with the characters of broad-spectrum anti-cancer and safe.But the mechanism of curcumin and NSCLC is not very clear.Many advanced patients have loose the opportunity of resection,with the poor tolerance of chemotherapy,and always failed treated because of the occurrence of resistance.Therefore it is important to search more safe,effective and pro-sensitive medicine for the treatment and prognosis of lung cancer patients. This research start from curcum ininhibit NSCLC proliferation and its mechanism,as well as the research of curcumin increase the sensitivity of gefitinib,intends to provide experimental evidences for curcumin treat NSCLC in clinical.MethodsThe research use A549 and SK-MES-1 as experimental support. MTS assay and colony forming assay detect the proliferation of the above two cell lines affect by curcumin; FCM observe curcumin’s effect on cell cycle, and detect the protein level of API and related cell cycle after treated curcumin by westernbloting.To discuss the effects of curcumin in proliferation and the related mechanism.Then use H1975(EGFR 1858R+T790M)ã€H3255(EGFRL858R) as experimental supports and MTS proliferation assay and Bliss method to calculate IC50.Compare the sensitivity of the above two cell lines to gefitinib with curcumin or not. Results1. After treated with different concentration of curcumin can decrease the ability of NSCLC cell lines A549 and SK-MES-1 in proliferationâ— Different concentration of curcumin can inhibit the proliferation of NSCLC cell lines, the effect is dose-dependence and time-dependence.â— Different concentration of curcumin can decrease the colony forming ability of NSCLC cell lines.â— Different concentration of curcumin can make A549 cell cycle arrest in G2/M phase,and didn’t make SK-MES-1 cell cycle arrest.â— Different concentration of curcumin influence the protein level of API, and the cell cycle related protein expression.2. Curcumin can inhibit the proliferation of EGFR mutation cell line H1975(EGFR L858R+T790M) and H3255(EGFRL858R)3. Curcumin can increase the sensitivity of H1975(EGFR L858R+T790M)ã€H3255(EGFR L858R)to gefitinib.ConclusionsCurcumin can inhibit proliferation of NSCLC cell lines through AP1 and cell cycle related proteins;Curcumin can increase the sensitivity of H1975 (EGFR L858R+T790M)ã€H355 (EGFR L858R) to gefitinib. |
