Objective:To explore the capacity of SDF-1α inducing endogenous cells homing, the root canals which embraced SDF-1α were implanted subcut-aneously into immune deficiency mice and maxillary models of canines respec-tively, then the specimens were observed by histological techniques. Through discussing the capacity of SDF-1α, we explore a new method for dental pulp regeneration without seed cells implanting.Methods:This study contains two experiments:Part 1:Extracted preomolars for orthodontic treatment were cut into 5mm root canal segments. The root canal segments were prepared by mechanical-chemical methods, and individed into two groups.The root canal segments of the experimental group were filled using 100ng/mL SDF-1α with adsorbing peptide hydrogel scaffold, and the control group with peptide hydrogel scaffold only.The root canal segments were implanted subcutaneously in 6-8 weeks old male nude mice for six weeks, all samples were stained with hematine-eosin.Part 2:Three canine aged 10-12 month were selected. After establishing the pulpectomy model,six mature anterior teeth of every canine were randomly divided into two groups. A total of 9 root canals of experimental group were filled with 100ng/mL SDF-la with adsorbing peptide hydrogel scaffold, and 9 root canals of the control group with peptide hydrogel scaffold only. Retrieved the teeth were stained with hematine-eosin (HE), and immunohistochemical staining was used to detect the expression of DSPP and CD34 in regenerative tissues after six weeks.Results1. The root segments were implanted subcutaneously shows that the red tissues were observed in the opening.HE staining shows that a large number of fibroblast-like cells and blood vessels were observed in the root canals of experimental group. There was no regeneration tissues with cells or blood vessels were observed in the control group.2. Retrieved the teeth from the canines shows that a large number of fibroblast-like cells and blood vessel were observed in the root canals of the experimental group teeth. There was no regeneration tissues with cells or blood vessels were observed in the control group.Immunohistochemical staining of DSPP and CD34 in the root canal of the experimental group were positive.Conclusion1.SDF-1α was potential to induce endogenous cells homing into the prepared root canals in subcutaneously hetetotopic model.2.SDF-1α can induce endogenous cells homing for dental-pulp-like tissue regeneration in pulpectomy model.3.SDF-1α can provide a new method for the regeneration of the dental pulp in no cell transplantation by inducing endogenous cells homing into the root canal.
