Objective Identification and genetic analysis of SNPs in promoter re--gulation and signal peptide region of omentin gene among Guangxi Han、 Zhuang、and Dong populations. Methods We identified SNPs in promoter regulation and signal peptide region of omentin gene among 206 Guangxi Han populations with normal glucose tolerance through DNA direct sequ-encing. Haploview software was used to analyse linkage disequilibrium a--nd calculate haplotypes.We compared the difference of SNPs distribution frequency among three ethnic groups (Han, Zhuang, Dong) with normal glucose tolerance, and analysed the correlation between type 2 diabetes a--nd SNPs. Results We got six SNPs loci in omentin gene promoter regu-lation region by DNA sequencing of Han populations with normal gluco--se tolerance:-31g/a,-36a/g,-82a/g,-171g/a,-260A/G and-269A/G,and one SNP locus in signal peptide region:Gly16Gly(A/G). The comparison am ong ethnic groups suggests that there is no-171g/a SNP in Zhuang group; All SNPs are in the same haploblock except-31g/a,-36a/g,,82a/g,-269’A/ G and Glyl6Gly(A/G) are in perfect linkagedisequilibrium (D’=1,r2=1) Each nationality has three haplotypes:Aaaga,Gggag and Aggag.The frequ-ency has no significant difference (P>0.05); There is a significant differe--nce between Zhuang and Dong,Dong and Han in-269A/G distribution fr--equency of genotype and allelomorph (P<0.05).The comparison between Han T2DM group and the control group shows that there is no significa--nt difference in genotype and allelomorph distribution frequency of tagS--NPs-36a/g,-171g/a,-260A/G and-269A/G (P>0.05). And there is no significant difference between two groups of three haplotypes:Aaaga,Ggg--ag and Aggag(P>0.05); Enzyme-linked sandwich immunoassay(ELISA) res--ults suggest that the serum level of omentin in T2DM group is lower th--an that in the control group(P<0.05).And the serum levels of omentin are different among three nationalities (P<0.05).Association analysis suggests SNPs in promoter regulation and signalpeptide region of omentin gene are not significantly associated with clinical variables in the the Han nationa--lity.Conclusion SNPs in promoter and signal peptide region of omentin gene in Guangxi Han populations are not associated with type 2 diabetes,it’ s not a risk factor of T2DM. There is a significant difference of-269A/G SNP distribution frequency of genotype and allelomorph between the Do--ng/Han、Dong/Zhuang.
