Objective: In this study, we established an animal model of non compression herniated intervertebral disctissue,testing the expression of interleukin-23 and interleukin-17 in serum at different time points in rats, To observe the morphological changes of nerve root inflammation,we further explored the mechanism of nerve root injury caused by non compression herniated intervertebral disctissue and provided a theoretical basis for clinical treatment of the disease.Methods : We choosed 48 male SD rats of October whose body weights are between 300-350 g. The rats were randomly divided into model group and control group which contained 24 rats.To observe the behavioral changes of rats, after Modeling 1 week、3 weeks and 5 weeks, the model group and the control group were randomly selected 8.And then,we determined the right hind paw retraction threshold. The expression of IL-17 and IL-23 in serum was detected by ELISA method. Morphological changes of L4、5 nerve roots were observed by HE staining.Results: 1、After modeling, the peak ofthe irritability and frequency behavior of the model group were in 3 weeks. When it comes to nearly 5 weeks,the behavior gradually returned to normal. While the control group did not appear hyperactivity and irritability of the situation. During the observation period, both groups of rats were free and their appetite was normal, which was no incontinence、bite limbs and mouth repeatedly licking hind paw phenomenon. 2、After modeling the 1 week and 3 weeks,comparison of model group and control group, the right hind paw withdrawal threshold decreased, while the expression of interleukin-23 and interleukin-17 in serum increased.The difference was statistically significant. After modeling5 weeks, the model group of the right hind paw shrinkage threshold, expression of interleukin-23 and interleukin-17 have returned to normal, compared with the control group. There were no significant differences between the two groups. 3、In the model group, the myelin sheath of the longitudinal section of the lumbar nerve root was disintegrated and showed the vacuole degeneration in the 1 week,3 weeks. The most serious injury is inthe group of 5 weeks. The control group after 1 weeks of lumbar nerve root are part of the longitudinal myelin disintegration and the 3 weeks returned to normal.Conclusions: 1 、 In the group of non compression herniated intervertebral disctissue, the early presence of IL-23/IL-17 inflammation axis mediated inflammatory reaction was at 3 weeks. In the group of 5 weeks,inflammatory immune response reached the peak. 2 、 IL-23/IL-17 inflammatory axis plays an important role in the nerve root injury of IL-23. IL-17 may increase the expression of LDH in LDH model mice, and provide a new target for the treatment of LDH.
