Objective: To investigate the lignan glycosides in Illigera rhodantha Hance.(LGIR) effect on collagen-induced arthritis in mice and its regulatory role in Th17 cell proliferation, which explore the mechanism of action. Methods: DBA/1 mice were used to establish the model of CIA, will make the successful model of DBA/1 mice divided into the CIA model group, different concentrations of LGIR groups, methotrexate(MTX) group and treatment these mice by irrigation stomach intervention, normal control group by irrigation stomach with physiological saline. After the intervention, the levels of serum IL-23 and IL-17 were detected in each group of mice, to observe the pathological changes of synovial membrane of the mice in each group, take the spleen and extract the lymphocytes, Flow Cytometry(FCM) was used to detect the expression of Th17 cells in different groups. Results: 1. CIA mouse model were successfully copied; 2. Compared with the model group, LGIR could improve the AI score of CIA mice, and the difference was statistically significant. 3. Compared with the model group, LGIR can improve the pathological score of the ankle joint in CIA mice, and the difference is statistically significant. 4. Compared with the model group, LGIR can regulate down the serum IL-23 and IL-17 levels, the difference was statistically significant. 5. The change of Th17 lymphocyte subset: compared with the control group, the proportion of Th17 cells of the model group was increased, which the difference is statistically significant. Compared with the CIA model group, the ratios of LGIR high dose group and the MTX group were lower than the model group, the difference between the groups has statistical significance. Conclusions: LGIR can improve local joint swelling of CIA mice, reduce the levels of serum IL-23 and IL-17 and reduce the degree of synovitis disease, especially the high dose group which can down-regulate the proportion of Th17 cell subset in lymphocyte of CIA mice. Therefore, we can infer that LGIR can suppress the activation of Th17 cell effectively.
