Objective: To explore the protective effect and the mechanism of bur dock root water extract on vascular endothelial cell injury. Methods: We used the lipopolysaccharide(LPS) to induce the damaged endothelial cell model, and randomly divided them into the blank control group, the mode l group, the group treated with low concentration, the group treated with median concentration, and the group treated with high concentration. Enzy me-linked immunosorbent assay(ELISA) was used to detect the expressio n levels of intercellular adhesion molecule-1(ICAM-1), tumor necrosis fac tor alpha(TNF-α), matrix metalloproteinase-1(MMP-1), and c-reactive protei n concentrations(hs-CRP) in the culture supernatant of cells. PCR was us ed to detect the expression levels of toll-like receptor 4(TLR4), nuclear f actor(NF-κB) inhibitory factor(I-κB) in cell. Results: The expression lev els of each inflammation factors TNF-α, MMP-1, ICAM-1, hs-CRP are lo wer in the group treated with drug compared to the model group(P<0.05). The expression levels of TLR4 and NF-κB are lower in the group tre ated with drug intervention compared to the injury model group(P<0.05).Conclusions: Burdock root water extract may probably inhibited the expre ssion of TLR4 and NF-κB in cells, thus inhibiting inflammatory cytokines,snch as TNF-α, MMP-1, ICAM-1, hs-CRP for protecting the cells.
