Th17/Treg Cells Imbalance And Intervention Study Of Acitretin In Patients With Psoriasis Vulgaris

Abstract/Summary:
Objective: To distinguish the difference of Thl7/Treg cells and related cytokines IL-17 and TGF-β1 between normal subjects and patients with psoriasis vulgaris, as well as changes before and after treatment with Acitretin. To explore the role of Thl7/Treg cells imbalance and the effect of Acitretinin patients with psoriasis pathogenesis.Method:1.30 cases of patients with moderate to severe psoriasis vulgaris were treated with oral Acitretin capsules for 8 weeks. Psoriasis Area and Severity Index scores were used to evaluate the treatment effect. 2.Flow cytometry was used to measure the Th17 and Treg cells percentage of CD4 + Treg cells in peripheral blood between experimental group and control group; 3.The levels of IL-17 and TGF-β1 secretion in serum were measured by Enzyme linked immunosorbent assay(ELISA); 4. 20 cases of healthy people were used as control group.Results: Compared with normal human controls, patients with psoriasis vulgaris displayed higher level of Th17 cells and IL-17 in peripheral blood, the ratios of TGF-β1 in serum were significantly lower than the control group, the difference was statistically significant(P<0.05), but there was no significant difference in the percentage of Treg cells and normal control group.After Acitretintreatment,the percentage of Th17 and IL-17 levels were statistically lower than before(P<0.05);And the percentage of Treg, TGF-β1 levels were increased evidentlycompared with before(P<0.05).At the same time, Compared with before PASI score decreased significantly after treatment with Acitretin(P<0.05). The total effective rate was86.67%. For patients before and after treatment,PASI score and the proportion of cells Th17, IL-17 levels were positively correlated, the TGF-β1 was negatively correlated(P<0.05) and with no correlation with Treg cells.Conclusion: The Th17 cells and IL-17 increasedin the peripheral blood of patients with psoriasis vulgaris,Treg cells remains unchanged, TGF-β1 decreased.Acitretincan reduce the number of Th17 cells and IL-17 levels and significantly increase the number of Treg cells and TGF-β1 levels.
