ObjectiveTo observate the effect of traditional Chinese medicine-jianpihuayu on the proliferation and invasion and migration of hepatocellular carcinoma cell SMMC-7721MethodsAccording to the serum pharmacology research principles, human hepatocellular carcinoma cell SMMC-7721 cells were vitro cultured with the serum of Wistar rat, who were given a gavage of JianpiHuayu or normal saline. The control group cells received 10% of normal serum cell culture medium, the experimental group was treated with containing 10%,20% medicated serum cell culture medium. Through CCK-8 colorimetric experiment, to estimate the cell survival rate (CV) and the inhibitory rate of cell proliferation of the two groups after 48 or 72hours’ culturing. Measure and calculate cell migration rate of The control group and the experimental group at 24h and 48h, in Wound Healing, to estimate the inhibitory effect of JianpiHuayu on cells migration that of human hepatocellular carcinoma cell SMMC-7721. To observe the inhibitory effect of JianpiHuayu on invasion of hepatocellular carcinoma cells SMMC-7721 through the Transwell experiment. invased cells of the two group on the PET film had been observed and calculated after 24h’s or 48h’s culturing. The experiment results were compared by "t" test, and the difference was statistically significant when P< 0.05.ResultsThe results of CCK-8:Calculated the cell inhibition rate of all experimental groups, according to the results of 48h,72h. The results showed that the inhibitory effect of JianpiHuayu on human hepatocellular carcinoma cell SMMC-7721 proliferation will be enhanced by extending the delivery time. The Wound Healing results indicated that the healing rate of cells of 20% medicated serum group can be significantly reduced in 24h. In 48h, the results showed that the cells healing rate of experimental group were both significantly decreased compared with control group. And the healing rate of 20%medicated serum group was more obviously decreased than the 10% medicated serum group. In Transwell invasive experiments after 48h administration the number of invased cells of 20% medicated group was decreased significantly compared with 20% normal serum group.GonclusionTraditional Chinese medicine-jianpihuayu has inhibitory effect on cell activity, cell proliferation and invasion and migration of human hepatocellular carcinoma cell SMMC-7721. And it has a certain time effect, and obvious concentration dependence.
