ObjectiveObservation by applying the method of tonifying kidney and strengthening essence of traditional Chinese medicine compound and astragaloside IV influence on systemic scleroderma fibroblasts, and by measuring the fibroblasts Collagen I, Fibronectin, Smad3, Flil and its mRNA to further elucidate the pathogenesis of scleroderma, explore scleroderma therapy new intervention targets, provide new ideas and ways for modern scleroderma therapy.MethodsIn vitro level, treated or untreated systemic scleroderma and normal fibroblast isolated from corresponding pairs were utilized to detect expression of collagen I and fibronectin Fibronectin, Smad3, Flil and its mRNA by Weston blot and quantitative real-time RT-PCR, and immunofluorescence technique to detect the activity of Smad3 and positioning of the regulation. scleroderma mouse model induced by bleomycin was used to evaluate the effects of the drug in vivo.Results1. Successfully established scleroderma mouse model:compared with normal control group, scleroderma CollagenI, Fibronectin mRNA in model group were increased (p<0.05); Compared with scleroderma model group, the method of invigorating the essence of traditional Chinese medicine compound, astragalus glycosides intervention of scleroderma mouse CollagenI and Fibronectin mRNA are reduced (p<0.05).2. Successful build scleroderma fibroblasts cell lines:compared with those of the NF SScF CollagenI and Fibronectin in mRNA were increased (ρ< 0.05); Compared with SScF,1,3,10,30 microns intervention of SScF CollagenI and Fibronectin mRNA were reduced (ρ<0.05).3. Compared with those of the NF SScF Flil mRNA expression is decreased obviously (ρ< 0.05);* and SScF, compared the different concentrations of astragalus armour glycosides intervention of SScF Flil mRNA rises (ρ<0.05).4. Compared with the normal fibroblasts, P-Smad3 increased significantly in scleroderma fibroblasts, astragalus glycosides intervention of scleroderma fibroblasts, P-Smad3 levels drop, inhibition of Smad3 into the nucleus and the combination of cofactors and activate the TGF-β gene transcription. conclusionMethod of tonifying kidney and strengthening essence of traditional Chinese medicine compound and astragaloside can attenuate fibrosis by inhibiting the TGF-β/Smad3/Flil axis insclerosis.
