The Differences Of Electroacupuncture At Different Acupoints On The Expression Of PGP9.5 In Mice Of Bowel Tissue With Function Constipation

Objective:The research used ice water irrigation stomach method to simulate the constipation-predominant functional constipation, investigating the effect differences of different acupuncture acupoints on functional constipation mice and the associated enteric nervous relationship, which further confirm the effect difference of acupuncture treatment on functional constipation and the part of the mechanism.Method:Select the100 CL57BL/6 wild-type mice in batches, First batch of 10 mice were isolated as normal group and the remaining 30 mice were modeled based on the wrong ice water irrigation stomach method.Used as observe the molding consequence.The second batch of 5 mice were isolated as normal group and the remaining 55 mice were repeat molding process.After the success of the molding, the mice were randomly divided into model, Shangjuxu, Tianshu, and Dachangshu groups with 5 mice in each group.All the groups of mice were placed in different cages and barbed wire were applied as the separator for dry and wet substances to collect feces. The changes in stool weight, propulsive rate of small intestine, and positive expression of PGP9.5 as intestinal neuronal marker of the mice before and after molding, electroacupuncture were recorded and analyzed.Results:1.The ice water irrigation stomach method can make mice faeces weight (wet weight and dry weight)and the propulsive rate of small intestine decrease(P<0.01/P<0.05).2.Model group of constipation mice of faeces weight and the small intestinal propulsion did not return to normal(P<0.05); The faeces weight and the propulsive rate of small intestine after acupuncture increased significantly. No difference between each acupuncture groups of the faeces weight..EA stimulation at "Shangjuxu"could recover the propulsive rate of small intestine of constipation mice.EA stimulation at"Shangjuxu"and "Tianshu"for recovery effect of propulsive rate of small intestine is statistically significant (P< 0.05).The faeces weight and the propulsive rate of small intestine of constipation mice without acupuncture treatment cound not fully recovered(P>0.05).3.The average optical density of positive expression of PGP9.5 in intestinal ganglion cells on jejunum and ileum of constipation mice decrease significantly(P<0.01).EA at "Shangjuxu, Tianshu, Dachangshu" the expression of jejunum segment increased significantly(P<0.01), there is no difference between each groups (P>0.05). EA at "Shangjuxu, Dachangshu" the expression of the ileum segment increased significantly(P<0.01).EA at "Tianshu" the expression of the ileum segment decrease significantly(P<0.01).4.Positive expression of PGP9.5 in intestinal ganglion cells in myenteric nerve plexus of model mice decrease, there had cavity change in intestinal ganglion cells.EA "Shangjuxu, Dachangshu "the positive expression increased and the cavity change better in jejunum and ileum.EA"Tianshu"positive expression in jejunum increased, but positive expression in ileum decreased, and the cavity change has no obvious change.Conclusion:1. The ice water irrigation stomach method can make mice faeces changed and lower quality. The propulsive rate of small intestine and positive expression of PGP9.5 in intestinal ganglion cells decrease, resulting in bowel dysfunction.2.EA at "Shangjuxu, Tianshu, Dachangshu" could make mice faeces weight and the propulsive rate of small intestine increase, and the promoting effect of the propulsive rate of small intestine propulsion rate, EA "Shangjuxu" is better than "Tianshu".3.The cause of effect difference between EA at "Shangjuxu" and "Tianshu" is EA at "Tianshu" could make the ganglion cells cavitation damage and the positive expression of PGP9.5 decreased in ileum.
