Objective:to study the San Huang decoction on MCF-7 tamoxifen resistance (MCF-7/tamR) cell oxidative stress level, the influence of the PI3K/AKT signal pathway and apoptosis, and preliminary discussion on its inner relationship.Methods:to establish a cell line MCF-7/tamR, CCK 8 method testing tamoxifen for breast cancer cells MCF-7, the influence of MCF-7/tamR proliferation, and detection of drug resistance index;While detecting San Huang decoction ofMCF-7/tamR proliferation. Immunofluorescence method to evaluate MCF-7, MCF-7/tamR and each dose group of ROS levels;Flow cytometry instrument for detecting various experimental cell apoptosis;Western Blot method to detect San Huang decoction of MCF-7/tamR apoptosis related proteins expression on the influence of detection at the same time San Huang decoction on MCF-7/tamR cell PI3K/AKT signal pathway of protein expression.Results:the successful build theMCF-7/tamR cell lines, cell morphology change, and of tamoxifen resistance ability has been enhanced, the RI index was 3.61±0.35 (p< 0.05), compared with wild typeMCF-7cell line, ROS levels, MCF-7/tamR cells apoptosis rate fell to 3.98% from 5.2%, and the Bcl-2 activity increased, Bax/Bak ratio is reduced, and PI3K/AKT pathway apparent activation, loss of PTEN expression. San Huang decoction of MCF-7/tamR cell lines have obvious inhibitory effect, IC50 alue concentration of 4.28±0.06 mg/mL, San Huang decoction can make MCF-7/tamR cells apoptosis rate increased to 6.46% and 8.95%, at the same time can cutBcl-2 activityof MCF-7/tamR, increase Bax/Bak,Can lower the level of ROS of drug resistant cell lines, inhibition of PI3K/AKT signal pathway, increase the expression of tumor suppressor gene PTEN.Conclusion:San Huang decoction can inhibit breast cancer MCF-7 tamoxifen, a drug resistance cell line (MCF-7/tamR) proliferation and promote its apoptosis, inhibiting ROS levels and inhibit the activity of PI3K/AKT signal pathway.The role of key may be through the inhibition of PI3K/AKT signaling pathway, thus inhibiting oxidative stress level.
